Micelle-like nanoparticles of PLA-PEG-PLA triblock copolymer as chemotherapeutic carrier.

Triblock copolymer PLA-PEG-PLA were synthesized using ring opening polymerization with different LA/EG ratio. Micellar aggregates were prepared from these block copolymers and characterized. The degradation characteristics of selected copolymers were assessed in both micellar and film forms. Surface segregation of PEG was also quantified as a function of copolymer composition. Anti-cancer drugs 5-FU and paclitaxel were loaded into the micellar nanospheres with good efficiency. The drug release profile showed good control over the release of paclitaxel from these polymers.

Drug loaded microfiber scaffolds have potential for sublingual drug delivery due to their fast dissolution time and tunable porosity. Such microfiber scaffolds can be prepared by melt electrowriting (MEW), wherein a polymer melt is electrostatically drawn out of a syringe onto a computer controlled moving collector. The fabrication of such scaffolds via MEW has previously been shown for a polymer with a glass transition temperature (T) just above room temperature, making handling challenging.

The physicochemical properties of emulsions based on poloxamers (triblock copolymers of a hydrophobic polyoxypropylene chain and two hydrophilic polyoxyethylene chains) depend on the composition and preparation method. This study examined the impact of poloxamer P188 concentration, autoclaving mode, heating, and salt presence on the viscosity, particle size distribution, and morphology of particles using viscometric analysis, dynamic light scattering (DLS), and atomic force microscopy (AFM). It was shown that sample preparation affects the particle size and morphology but not the chemical composition of P188.

Introduction of non-DLVO forces by nonionic surfactants brings about fascinating changes in the phase behavior of silica nanosuspensions. We show here that alterations in the interaction and wetting properties of negatively charged silica nanoparticles (Ludox® LS) in the presence of polyethylene oxide-polypropylene oxide-polyethylene oxide-based triblock copolymers called Pluronics lead to the formation of stable o/w Pickering emulsions and interparticle attraction-induced thermoresponsive liquid-liquid phase separations. The results make interesting comparisons with those reported for Ludox® TM nanosuspensions comprising larger silica nanoparticles.

Poly(-lysine)--poly(ethylene glycol)--poly(-lysine) triblock copolymers for the preparation of flower micelles and their irreversible hydrogel formation.

Poly(-lysine)--poly(ethylene glycol)--poly(-lysine) (PLys--PEG--PLys) triblock copolymers formed polyion complex (PIC) with poly(acrylic acid) (PAAc) or sodium poly(styrenesulfonate) (PSS), leading to the formation of flower micelle-type nanoparticles (Nano or Nano) with tens of nanometers size in water at a polymer concentration of 10 mg/mL. The flower micelles exhibited irreversible temperature-driven sol-gel transitions at physiological ionic strength, even at low polymer concentrations such as 40 mg/mL, making them promising candidates for injectable hydrogel applications. Rheological studies showed that the chain length of PLys segments and the choice of polyanions significantly impacted irreversible hydrogel formation, with PSS being superior to PAAc for the formation.
