AI Article Synopsis

  • PTTG is a gene that resembles securin and is important in cell transformation, but its role in normal liver tissues is not well understood, particularly during liver regeneration.
  • PTTG expression in rat liver cells increased with hepatocyte growth factor (HGF) and decreased with transforming growth factor-beta1, indicating its involvement in liver cell proliferation.
  • The study suggests that PTTG could serve as a new marker for tracking liver cell proliferation during regeneration processes in both laboratory and living subjects.

Article Abstract

Background: Pituitary tumor transforming gene (PTTG), homologous to a mammalian securin, plays a pivotal role in cell transformation, however, its biological function(s) in normal tissues is not fully understood. Because the liver is a regenerative organ, the relevant biological function of PTTG in the liver would be more feasible to understand PTTG. Also, PTTG may be involved in the liver regeneration.

Materials And Methods: Expressions of rat hepatic PTTG messengerRNA (mRNA) and cellular immunoreactivities during the cell proliferative period of the liver regeneration both in vitro and in vivo were tested.

Results: PTTG expression of the rat primary hepatocyte was stimulated by HGF in a dose dependent manner, and was suppressed when hepatocyte proliferation was inhibited by transforming growth factor-beta1. A positive PTTG immunoreactive co-localizing with 5-bromo-2'-deoxyuridine (BrdU) in the hepatocyte nucleus was found and there was a concurrent sister chromatin itself by the immunofluorescent labeling of PTTG with cytokeratin 18 (CK18).

Discussion: Since the correlation of PTTG mRNA expression, cell proliferation and immunoreactivity were observed in primary rat cultured hepatocytes, PTTG may be a novel marker of cell proliferation both in vitro and in vivo liver regeneration.

In this study, we designed a fusion protein, rhCR, by combining human collagen with the self-assembling peptide RADA-16 using genetic engineering technology. The rhCR protein was successfully expressed in . The rhCR can self-assemble into a three-dimensional nanofiber network under physiological conditions.

Liver tissue engineering holds promising in synthesizing or regenerating livers, while the design of functional scaffold remains a challenge. Owing to the intricate simulation of extracellular matrix structure and performance, porous scaffolds have demonstrated advantages in creating liver microstructures and sustaining liver functions. Currently, various methods and processes have been employed to fabricate porous scaffolds, manipulating the properties and morphologies of materials to confer them with unique supportive functions.

Hepatocytes are organized into distinct zonal subsets across the liver lobule, yet their contributions to liver homeostasis and regeneration remain controversial. Here, we developed multiple genetic lineage-tracing mouse models to systematically address this. We found that the liver lobule can be divided into two major zonal and molecular hepatocyte populations marked by Cyp2e1 or Gls2.

Although hemostatic powders are commonly used in clinical and emergency settings, they frequently show poor absorption, raise cytotoxicity issues, and are not effective for fatal non-compressible bleeding. The purpose of this research is to create a self-gelling hemostatic powder based on chitosan, bentonite, and sodium polyacrylate (CBS) to improve the hemostatic effect. When liquid comes into contact with CBS powders, they can fuse and form a stable hydrogel in less than 30s.
