[The role of circadian gene period1 in morphine reward in mice].

Objective: To investigate the role of circadian gene Period1 on drug dependence.

Method: The ribozyme cleave the Period1 mRNA (per1RZ) was designed and the per1RZ expression vector (pcDNA 3.1-per1RZ) was constructed based on eukaryotic expression vector pcDNA 3.1. In vitro cleavage experiment proved the target cleave Period1 mRNA ability of per1RZ. Conditional place preference (CPP) paradigm used to investigate the effect of intracerebroventricular (ICV) injection of pcDNA 3.1-per1RZ on drug reward in BALB/C mice.

Result: Quantitative analysis of in vitro cleavage experiment showed the efficacy of ribozyme per1RZ, about 60% of the Period1 mRNA was cleaved by ribozyme per1RZ. CPP test displayed the block of drug reward in pcDNA 3.1-per1RZ ICV injection group. Period1 expression in brain was attenuated of pcDNA 3.1-per1RZ ICV injection group demonstrated by western blot.

Conclusion: Interfere the Period1 expression in brain could attenuate the psychological dependence of drug in mammals.

In our research, we constructed models of renal ischemia-reperfusion (I/R)-exposed acute kidney injury (AKI) and unilateral ureteral obstruction (UUO)-stimulated renal fibrosis (RF) in C57BL/6 mice and HK-2 cells. We firstly authenticated that oral pinocembrin (PIN) administration obviously mitigated tissue damage and renal dysfunction induced by I/R injury, and PIN attenuated UUO-caused RF, as confirmed by the reduced expression of fibrotic markers as well as hematoxylin-eosin (H&E), Sirius red, immunohistochemistry, and Masson staining. Meanwhile, the beneficial role of PIN was again demonstrated in HK-2 cells with hypoxia-reoxygenation (H/R) or transforming growth factor beta-1 (TGF-β1) treatment.

To investigate the effects of long non-coding RNA KLHL7-AS1 (LncRNA KLHL7-AS1) on the proliferation and apoptosis of nucleus pulposus cells under oxidative stress and its mechanisms. Human nucleus pulposus cells (HUM-iCell-s012) were divided into 4 groups, and unoxidized nucleus pulposus cells were transfected with an empty pcDNA vector (pcDNA-control) to serve as the blank control group. Based on previous studies on oxidative stress-induced nucleus pulposus cell senescence and preliminary experiments, oxidative stress was induced by treating nucleus pulposus cells with 400 μmol/L HO.

Canids act as a crucial intermediary in the transmission of rabies and , serving as co-infection hosts and pathogen carriers for both rabies and hydatid disease (HD) transmitted from animals to humans. Therefore, an effective and efficient bivalent oral vaccine for preventing HD and rabies is urgently required to reduce economic losses in husbandry resulting from rabies and HD. In this study, a full-length plasmid (pcDNA4-NPM+G+EgM123+eGFP+L) carrying the gene and fluorescence reporter genes of eGFP and four auxiliary transfection plasmids of rabies virus SRV (pcDNA4-N, pcDNA4-P, pcDNA4-G, pcDNA-L) were established by reverse genetics approaches and co-transfected to BSR cells by electrotransfection.

Developing human papillomavirus (HPV) therapeutic DNA vaccines requires an effective delivery system, such as cell-penetrating peptides (CPPs). In the current study, the multiepitope DNA constructs harboring the immunogenic and conserved epitopes of the L1, L2, and E7 proteins of HPV16/18 (pcDNA-L1-L2-E7 and pEGFP-L1-L2-E7) were delivered using KALA and REV CPPs with different properties in vitro and in vivo. Herein, after confirmation of the REV/DNA and KALA/DNA complexes, their stability was investigated against DNase I and serum protease.
