Objectives: To investigate the effect of short interfering RNA targeting MAT 2A on growth and apoptosis of hepatoma cells.

Methods: The four siRNA against MAT 2A gene were transcript synthesized intracelluarly by expressed templates of plasmid vector pSilence-2.1-U6. We inserted the target sequence of MAT 2A gene into the upstream of the reporter gene in order to construct the recombinant plasmid vector plucA-MAT 2A. The recombinant plasmid and siRNA-producing plasmid were co-transfected into 293 T cells using this construct via lipofectamine methods. The inhibition effect was detected by measuring luciferase activity in the cell lysate to screen the effective siRNA, and then, the effective siRNA was transfected into Bel-7402 cells. The effect of siRNA treatment on the MAT 2A mRNA level and the MAT activity of hepatoma cells were measured. In order to study the effect of short interfering RNA targeting MAT 2A on growth and apoptosis of hepatoma cells, the tumor cell killing rate was analyzed by MTT method and the rate of apoptosis of hepatoma cells was evaluated by flow cytometry.

Results: The two siRNA among the four siRNA displayed inhibitory effect on the lucifermase expression with the inhibitory rates of 81% and 89% respectively. The expression of MAT 2A mRNA in Bel-7402 cells was specifically inhibited and the MAT activity in Bel-7402 cells was decreased. Furthermore, silencing of the MAT 2A gene by RNAi significantly inhibited hepatoma cell growth and led to induction of apoptosis.

Conclusion: RNA interference-mediated silencing of MAT 2A gene attenuates growth and induces apoptosis of hepatoma cells; MAT 2A is an ideal target of gene-specific therapy for liver cancer.

The methylotrophic yeast belongs to the group of homothallic fungi that are able to spontaneously change their mating type by inversion of chromosomal DNA in the MAT locus region. As a result, natural and genetically engineered cultures of these yeasts typically contain a mixture of sexually dimorphic cells that are prone to self-diploidisation and spore formation accompanied by genetic rearrangements. These characteristics pose a significant challenge to the development of genetically stable producers for industrial use.

Genomic analysis of mobility measures on 5-month-old gilts associated with structural soundness.

J Anim Sci

USDA-Agricultural Research Service, U.S. Meat Animal Research Center (USMARC), Clay Center, NE, USA.

Sow lameness results in premature culling, causing economic loss and well-being issues. A study, utilizing a pressure-sensing mat (GAIT4) and video monitoring system (NUtrack), was conducted to identify objective measurements on gilts that are predictive of future lameness. Gilts (N = 3656) were categorized to describe their lifetime soundness: SOUND, retained for breeding with no detected mobility issues; LAME_SOW, retained for breeding and detected lame as a sow; CULL_STR, not retained due to poor leg structure; LAME_GILT, not retained due to visible signs of lameness; and CULL, not retained due to reasons other than leg structure.

To date, only a few microbial community studies of cold seeps at the South China Sea (SCS) have been reported. The cold seep dominated by tubeworms was discovered at South Yungan East Ridge (SYER) offshore southwestern Taiwan by miniROV. The tubeworms were identified and proposed as sp.

Objective: Triple-Negative Breast Cancer (TNBC), the most challenging subtype of Breast Cancer (BC), currently lacks targeted therapy, presenting a significant therapeutic gap in its management. Tumor Associated Macrophages (TAMs) play a significant role in TNBC progression and could be targeted by repolarizing them from M2 to M1 phenotype. Matairesinol (MAT), a plant lignan, has been shown to exhibit anticancer, anti-inflammatory and immunomodulatory activities.

Utilizing machine learning to optimize agricultural inputs for improved rice production benefits.

Crop Ecophysiology and Cultivation Key Laboratory of Sichuan Province, Rice Research Institute / State Key Laboratory of Crop Gene Exploration and Utilization in Southwest China, Sichuan Agricultural University, Chengdu 611130, China.

Lower efficiency of agricultural inputs in the four conventional rice planting methods limits productivity and environmental benefits in Southwest China. Thus, we developed a machine-learning-based decision-making system for achieving optimal comprehensive benefits during rice production. Based on conventional benefits for achieving optimal benefits, implemented strategies in these planting methods: reducing N fertilizer by 16% while increasing seed inputs by 9% in mechanical transplanting (MT) method improved yield and environmental benefits; reducing N fertilizer and seed inputs by 10-12% in mechanical direct seeding (MD) method decreased environmental impacts; increasing N-K fertilizers and seed inputs by 15-33% in manual transplanting (MAT) method improved its comprehensive benefits by 7-14%; applying N-P-K fertilizer ratio of 2:1:2 in manual direct seeding (MAD) method enhanced yield.
