The reaction between 1,3-bis(3,5-dimethylpyrazolylmethyl)hexahydropyrimidine L and Mo(CO) in CHCN at 130 °C afforded a binuclear Mo(0) complex 1 containing a new macrocycle formed upon C-N bond cleavage in L in good yield. Conversely, a clean reaction takes place between L and [Mo(CO)(COD)] in THF at 60 °C to give a new metalloligand complex [Mo(CO)(κ-,-L)] 2 containing a spectator pyrazole arm in 83% yield. Their structures were determined by X-ray diffraction methods, and a plausible mechanism is proposed for the C-N bond cleavage leading to complex 1.

Pharmacological inhibition of the MAP2K7 kinase in human disease.

Front Oncol

December 2024

Department of Pathology and Immunology, Baylor College of Medicine, Houston, TX, United States.

The MAP2K7 signaling pathway activates the c-Jun NH2-terminal protein kinase (JNK) in response to stress signals, such as inflammatory cytokines, osmotic stress, or genomic damage. While there has been interest in inhibiting JNK due to its involvement in inflammatory processes and cancer, there is increasing focus on developing MAP2K7 inhibitors to enhance specificity when MAP2K7 activation is associated with disease progression. Despite some progress, further research is needed to fully comprehend the role of MAP2K7 in cancer and assess the potential use of kinase inhibitors in cancer therapy.

Electrochemical Oxidative Cascade Cyclization of Alkenyl Alcohols with External Nucleophiles to Access Amino- and Hydroxy-Functionalized -Heterocycles.

Key Laboratory of Biocatalysis & Chiral Drug Synthesis of Guizhou Province, School of Pharmacy, Zunyi Medical University, Zunyi 563003, P. R. China.

A convenient electrochemical oxidative cascade cyclization of alkenes equipped with pendant alcohols with general nucleophiles was developed. Using readily available diarylmethanimine and carboxylic acids as nucleophilic sources, a broad range of internal alkene and terminal alkene substrates could produce RCO- and ArCN-functionalized -heterocycles in moderate to high yields without the requirement for external oxidants and metals. These resulting products can subsequently be hydrolyzed to yield valuable NH- and OH-functionalized tetrahydrofurans and tetrahydropyranes under mild conditions.

Article Synopsis
  • Auditory neuropathy (AN) is a type of hearing loss caused by damage to inner hair cells, ribbon synapses, and spiral ganglion neurons.
  • The lack of effective treatments has led to a need for better understanding of how AN develops, particularly through the role of MAPK signaling.
  • Research shows that changes in various MAPK pathways, specifically ERK, p38, and JNK, occur at different sites in the auditory system, indicating that targeting these pathways could help in developing new treatments for AN.
