Nitroaromatic compounds are used extensively in many industrial processes and have been released into the environment where they are considered environmental pollutants. Nitroaromatic compounds, in general, are resistant to oxidative attack due to the electron-withdrawing nature of the nitro groups and the stability of the benzene ring. However, the bacterium Comamonas sp. strain JS765 can grow with nitrobenzene as a sole source of carbon, nitrogen and energy. Biodegradation is initiated by the nitrobenzene dioxygenase (NBDO) system. We have determined the structure of NBDO, which has a hetero-hexameric structure similar to that of several other Rieske non-heme iron dioxygenases. The catalytic subunit contains a Rieske iron-sulfur center and an active-site mononuclear iron atom. The structures of complexes with substrates nitrobenzene and 3-nitrotoluene reveal the structural basis for its activity with nitroarenes. The substrate pocket contains an asparagine residue that forms a hydrogen bond to the nitro-group of the substrate, and orients the substrate in relation to the active-site mononuclear iron atom, positioning the molecule for oxidation at the nitro-substituted carbon.

Article Synopsis
  • Researchers have isolated a bacteria, Raoultella ornithinolytica CT3, from silkworm excrement that can effectively break down the antibiotic chloramphenicol (CAP) in just 48 hours.
  • The study identified 22 metabolites produced during CAP degradation and established that the process follows first-order kinetics, mainly involving acetylation and other auxiliary pathways.
  • Genomic analysis revealed specific genes related to CAP breakdown, and while the intermediate metabolites were slightly toxic, the final products were found to be environmentally safe, contributing to understanding CAP's microbial degradation.

Rieske oxygenases are known as catalysts that enable the cleavage of aromatic and aliphatic C-H bonds in structurally diverse biomolecules and recalcitrant organic environmental pollutants through substrate oxygenations and oxidative heteroatom dealkylations. Yet, the unproductive O activation, which is concomitant with the release of reactive oxygen species (ROS), is typically not taken into account when characterizing Rieske oxygenase function. Even if considered an undesired side reaction, this O uncoupling allows for studying active site perturbations, enzyme mechanisms, and how enzymes evolve as environmental microorganisms adapt their substrates to alternative carbon and energy sources.

Article Synopsis
  • Rieske non-heme dioxygenase enzymes are critical for breaking down nitroaromatic pollutants, but many harmful compounds like 2,4-dichloronitrobenzene (24DCNB) lack effective natural enzymes for degradation.
  • Researchers engineered a dioxygenase from a specific bacterial strain using molecular simulations and targeted mutations, achieving a significant improvement in its activity toward 24DCNB—up to a 62-fold increase.
  • This study demonstrates how computational methods can be used to enhance enzyme functions, aiding in the biodegradation of challenging environmental pollutants.

Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by eczema-like skin lesions, dry skin, severe itching, and recurrent recurrence. The whey acidic protein four-disulfide core domain gene WFDC12 is highly expressed in skin tissue and up-regulated in the skin lesions of AD patients, but its role and relevant mechanism in AD pathogenesis have not been studied yet. In this study, we found that the expression of WFDC12 was closely related to clinical symptoms of AD and the severity of AD-like lesions induced by DNFB in transgenic mice.

Improvement of refractory nitrogen-containing organics biodegradation is crucial to meet discharged nitrogen standards and guarantee aquatic ecology safety. Although electrostimulation accelerates organic nitrogen pollutants amination, it remains uncertain how to strengthen ammonification of the amination products. This study demonstrated that ammonification was remarkably facilitated under micro-aerobic conditions through the degradation of aniline, an amination product of nitrobenzene, using an electrogenic respiration system.
