The release of holocytochrome c (cyt c) from mitochondria into the cytosol is reportedly a landmark of the execution phase of apoptosis. As shown here, the P-glycoprotein- (P-gp) expressing K562/ADR cell line (but not the parental K562 cell line) exhibits both cytosolic and mitochondrial cyt c in the absence of any signs of apoptosis. K562/ADR cells were found to be relatively resistant to a variety of different inducers of apoptosis, and blocking the P-gp did not reverse this resistance. The release of cyt c in non-apoptotic K562/ADR cells was not accompanied by that of any other mitochondrial apoptogenic protein, such as AIF or Smac/DIABLO, and was inhibited by Bcl-2 over expression. In addition, using a cell-free system, we show that mitochondria isolated from K562/ADR cells spontaneously released cyt c. These data suggest that cyt c release may be compatible with the preservation of mitochondrial integrity and function, as well as cell proliferation.

Article Synopsis
  • The study aimed to explore how asiatic acid (AA) affects the drug resistance in human leukemia cells (K562/ADR) resistant to adriamycin (ADR).
  • AA was found to reduce the resistance of these cells and enhance the effectiveness of ADR, as shown by various assays including CCK-8 and flow cytometry.
  • The results indicated that AA down-regulates the expression of certain proteins related to drug resistance, suggesting a potential mechanism for reversing resistance in these cancer cells.

Drug-induced self-assembled nanovesicles for chloroquine to sensitize MDR tumors to mitoxantrone hydrochloride.

Colloids Surf B Biointerfaces

January 2025

Ministry of Educational (MOE) Key Laboratory of Macromolecular Synthesis and Functionalization, Department of Polymer Science and Engineering, Zhejiang University, Hangzhou 310058, China. Electronic address:

Multidrug resistance (MDR) is an incidental trouble post-chemotherapy, necessitating innovative therapeutic strategies. This study explores the potential of chloroquine (CQ) as a sensitizer for mitoxantrone hydrochloride (MitH) in drug-resistant tumors and introduces a novel pH-responsive drug-induced self-assembly nanovesicle (DIV) based on an amphiphilic polyphosphonitrile (PPAP) for the co-delivery of MitH and CQ. PPAP cannot self-assemble into nanovesicles alone, but when a certain amount of MitH was added, the multiple non-covalent interactions between PPAP and MitH contributed to the formation of DIV, which exactly improved the co-loading content of MitH and CQ to a large extent.

Introduction: One of the many reasons for cancer treatment failure and recurrence is acquired Multidrug Resistance (MDR). Overcoming cancer drug resistance has been the focus of researchers' studies. Cellular prion protein (PrP) is a glycophosphatidylinositol-anchored cell-surface glycoprotein that has been implicated in tumor behavior, including proliferation, apoptosis, invasion, metastasis, and chemoresistance.

Cytotoxic and anti-inflammatory polyacetylenes from Tridax procumbens L.

Phytochemistry

September 2024

School of Pharmacy, Guangdong Pharmaceutical University, Guangzhou, 510006, China; Guangdong Engineering Research Center for Lead Compounds & Drug Discovery, Guangzhou, 510006, China. Electronic address:

Herein, 17 previously undescribed polyacetylenes and 9 known ones were isolated from Tridax procumbens L. Their structures were identified using spectroscopic techniques (NMR, UV, IR, MS and optical rotation), the modified Mosher method, electronic circular dichroism (ECD) data and ECD calculation. The cytotoxicity of polyacetylenes on six human tumour cell lines (K562, K562/ADR, AGS, MGC-803, SPC-A-1 and MDA-MB-231) was evaluated.

Article Synopsis
  • The study aimed to explore how gene expression relates to the prognosis of acute myeloid leukemia (AML) patients by analyzing bone marrow cells from 27 individuals and comparing results with healthy subjects and different AML patient groups.
  • Results show that patients with refractory AML had significantly higher levels of a specific gene compared to those responsive to chemotherapy, and those with extramedullary infiltration also exhibited increased gene expression.
  • Analysis revealed that resistant leukemia cell lines expressed more of the gene at both mRNA and protein levels compared to sensitive cell lines, suggesting a potential link between this gene and treatment resistance in AML.
