The use of reporter genes such as beta-galactosidase and green fluorescent protein is a powerful molecular tool for the visualization of in-vivo gene expression. In this paper, we describe the generation of mice that possess a beta-galactosidase reporter gene introduced into the endothelin-B receptor locus by homologous recombination in embryonic stem cells. These mice express beta- galactosidase wherever endothelin-B is expressed and therefore provide a precise in-vivo localization profile of endothelin-B expression.

The endothelin-B receptor (ETR) mediates vasodilation in young women, an effect that is absent in postmenopausal women. We have previously demonstrated that ETR-mediated vasodilation is regulated by estradiol (E) in young women; however, the impact of E on ETR function in postmenopausal women remains unknown. Accordingly, the objective of this study was to test the hypothesis that E exposure restores ETR-mediated dilation in postmenopausal women.

Two major ligand-receptor signaling axes - endothelin Edn3 and its receptor Ednrb, and glial-derived neurotrophic factor (GDNF) and its receptor Ret - are required for migration of enteric nervous system (ENS) progenitors to the hindgut. Mutations in either component cause colonic aganglionosis, also called Hirschsprung disease. Here, we have used Wnt1Cre and Pax2Cre in mice to show that these driver lines label distinct ENS lineages during progenitor migration and in their terminal hindgut fates.

Methods: LPS or saline was injected intraperitoneally in KO (KO-LPS; n = 9, KO-sal; n = 5) and wild-type (WT) (WT-LPS; n = 6, WT-sal; n = 6) pups obtained within 24 h of birth.
