Background: Neurofilament Light Chain (NfL) is a blood biomarker of axonal injury and neurodegeneration that can be used in a variety of neurological disorders. Despite the potential clinical use of plasma NfL across multiple neurological disorders, there is increasing evidence that underlying comorbidities such as renal impairment associated with chronic kidney disease (CKD) and cardiovascular diseases can increase NfL concentrations. Background: Brain aging is a complex process influenced by various genetic, lifestyle, and environmental factors, as well as by age-related and often co-existing pathologies. MRI and AI methods have been instrumental in understanding neuroanatomical changes that occur during aging in large and diverse populations.

Method: We leveraged Surreal-GAN, a state-of-the-art deep representation learning method, to elucidate the heterogeneity of brain aging by deriving dominant dimensions (patterns) of brain changes and examining their associations with various clinical and genetic factors.

Background: Dementia patients often have several co-existing diseases, whereas the specific temporality and development patterns between them remain uncertain.

Method: Based on the multicenter, community-based prospective UK Biobank, enriched disease diagnoses were extracted from hospital admission linkage, along with basic demographic information and individual genome data from the baseline visit. We constructed disease trajectory before dementia utilizing the phenome-wide association analysis to firstly filter potential precursors, the directional test to select temporal disease pairs, and conditional logistic regression to finally quantify association strength.

Background: Vascular contributions to cognitive impairment and dementia (VCID) results from cerebrovascular injuries, significantly contributing to age-related cognitive decline, and coexists with Alzheimer's disease (AD) in excess of 70% of AD patients. These co-occurring neuropathological subtypes are referred to as mixed-etiology dementia (MED). Despite the prevalence of MED little is known regarding the neuroinflammatory responses of microglia in the context of vascular injury in tissues already containing AD-related cerebral amyloidosis.
