High-throughput screening techniques continue to provide important tools to the pharmaceutical industry for the efficient identification of drug leads. However, high-throughput techniques are now being exploited to address a variety of pharmacological and cellular signaling research questions, including the regulation and role of intracellular Ca(2+) in a plethora of biological systems. Although an awareness of specific assay conditions is crucial for reliable and reproducible measurements of intracellular free Ca(2+) whichever system of study is used, the complex temporal nature of Ca(2+) signals has posed some unique limitations for its measurement in high-throughput mode. Progress in high-throughput design has overcome many of these limitations and will complement other technical approaches to understanding the underlying regulation and role of intracellular Ca(2+).

Transient receptor potential melastatin-4 (TRPM4) ion channel expression is upregulated in prostate cancer (PCa), contributing to increased cell proliferation, migration, adhesion, epithelial-to-mesenchymal transition, cell cycle shift, and alterations of intracellular Ca signaling. GEO2R platform analysis of messenger RNA (mRNA) expression of ~ 6350 genes in normal and malignant prostate tissue samples from 15 PCa patients demonstrates that TRPM4 expression is upregulated sixfold and is among the most significantly upregulated genes in PCa. We find that absence of TRPM4 reduced PCa tumor spheroid size and decreased PCa tumor spheroid outgrowth.

Objectives: To investigate the role of the BNIP3-PI3K/Akt signaling pathway in mediating the inhibitory effect of Decoction (BYHWT) on mitochondrial autophagy in human synovial fibroblasts from rheumatoid arthritis patients (FLS-RA) cultured under a hypoxic condition.

Methods: Forty normal Wistar rats were randomized into two groups (=20) for daily gavage of BYHWT or distilled water for 7 days to prepare BYHWT-medicated or control sera. FLS-RA were cultured in routine condition or exposed to hypoxia (10% O) for 24 h wigh subsequent treatment with IL-1β, followed by treatment with diluted BYHWT-medicated serum (5%, 10% and 20%) or control serum.
