The permeability of the pineal gland to lanthanum was investigated by means of transmission electron microscopy and X-ray microanalysis. Different pineal cell populations were studied. Light pinealocytes as well as glial cells were not permeated by the tracer lanthanum, which normally stays extracellularly. Dark pinealocytes showing different degrees of degenerative changes are permeated by lanthanum. The loss of plasma membrane integrity explains the entry of lanthanum into these cells. We conclude: (1) the lanthanum tracer can be used as an indicator of an alteration in membrane permeability, (2) dark pinealocytes appear as cells in the state of degeneration and represent a different physiological state than light pinealocytes.

Background: The epiphysis cerebri (pineal gland) is a small-sized, photo neuroendocrine organ in the brain of most vertebrates. Their effect is through secretion of melatonin, a serotonin-derived hormone which is stimulated by darkness and inhibited by light and modulates the circadian rhythm; light and dark cycle like a biological clock, sleep patterns (sleep-wake cycle), and sexual development.
Aim: This study aimed to identify and differentiate the different cell types filling the pineal gland parenchyma of mature male sheep.

Melatonin is an endogenous indolamine, synthesized and secreted from the pineal gland. The environmental light-dark cycle is the primary regulator of melatonin synthesis. Darkness during the subjective night induces noradrenaline secretion, which stimulates pinealocytes for melatonin production.
