The absolute configuration assignments of three antifungal agents, (+)-(2R,4S)-ketoconazole, (+)-(2R,4S)-itraconazole (with (S)-configuration at the sec-butyl group) and (+)-(S)-miconazole nitrate have been confirmed by using vibrational circular dichroism (VCD). For these three antifungal drugs, this study also provides evidence for the most abundant conformations of miconazole and for the relative conformations of the azole, dichlorophenyl, and methoxyphenyl groups in ketoconazole and itraconazole, in chloroform solution.

Contrast transfer function (CTF) estimation is essential to the data processing workflow of cryo-electron tomography (cryoET). Here, we present a protocol for CTF estimation of the cryoET tilt series with CTFMeasure. CTFMeasure can estimate the CTF parameters together with the absolute tilt angle offset of the sample.

Significance: In the last years, time-resolved near-infrared spectroscopy (TD-NIRS) has gained increasing interest as a tool for studying tissue spectroscopy with commercial devices. Although it provides much more information than its continuous wave counterpart, accurate models interpreting the measured raw data in real time are still lacking.
Aim: We introduce an analytical model that can be integrated and used in TD-NIRS data processing software and toolkits in real time.

Chirality plays a crucial role in the pharmacological activity of triazoles, a key scaffold in antifungal agents and various therapeutic applications. This study focuses on optimizing the enantiomeric resolution of chiral triazoles using supercritical fluid chromatography (SFC) and 10 different columns, either immobilized or coated, chlorinated or nonchlorinated, cellulose or amylose-based chiral stationary phases (CSPs). Four novel triazoles and two marketed ones (tebuconazole and hexaconazole) were separated to determine optimal resolution conditions.

Nine undescribed prenylxanthone derivatives, aspergixanthones L-T (1-9), were isolated from the fungus Aspergillus stellatus. Compound 4 represents the first propionylated prenylxanthone, while compounds 7-9 are the first examples of prenylxanthones adorned with a 2,3-butanediol group. Their structures with absolute configurations were elucidated based on comprehensive spectroscopic analyses, ECD calculation, single-crystal X-ray diffraction, Mo(OAc) induced ECD experiment, and Mosher's method.
