Oncolytic herpes simplex viruses (oHSV) preferentially replicate in cancer cells while inducing antitumor immunity, and thus, they are often referred to as in situ cancer vaccines. OHSV infection of tumors elicits diverse host immune responses comprising both innate and adaptive components. The sensitivity of human glioblastoma cells to virus-mediated oncolysis was investigated on five patient-derived cell lines. Primary glioblastoma cells (Gbl13n, Gbl16n, Gbl17n, Gbl25n, and Gbl27n) were infected with tenfold serial dilutions of the Leningrad-3 strain of the mumps virus, and virus reproduction and cytotoxicity were monitored for 96-120 h. Immortalized human non-tumor NKE cells were used as controls to determine the virus specificity.

Cytomegalovirus (CMV) infects a wide range of cell types, including tumor-associated myeloid cells and glioma cells. Clinical observations suggest a potential link between long-term glioblastoma survival and CMV reactivation. We herein present an oncolytic CMV vector, AD169r, which includes a restored pentamer complex gH/gL/pUL128-131 and the removal of UL1-UL20 and UL/b' sequences.

Molluscum contagiosum (MC) virus is a poxvirus that infects epidermal keratinocytes producing cutaneous nodules with characteristic intracytoplasmic inclusions. Intradermal nevus is a benign lesion that typically presents as clusters of melanocytes within the dermal layer of skin. Although both MC and melanocytic nevus are common lesions, MC arising within a melanocytic nevus is a rare event with only a few cases published in the literature.
