A highly unsaturated new amide, (2E,4Z,8Z,10Z)-N-isobutyl-2,4,8,10-dodecatetraenamide (1), was isolated in very small quantities from the whole plant of Asarum forbesii Maxim. together with four known compounds, (2E,4E,8Z,10E)-N-isobutyl-2,4,8,10-dodecatetraenamide (2), (-)-sesamin (3), (-)-asarinin (4) and (E)-asarone (5). The Z/E isomers, 1 and 2, were separated successfully by developed silver-ion medium-pressure liquid chromatography (SIMPLC). Compound 2 and the two diastereoisomers, 3 and 4, were isolated from this plant for the first time. The characterization of these compounds was achieved by various spectroscopic methods.

Recorded in Da he wai ke (The External Medicine of Da He) in the Ming dynasty without a corresponding disease title in contemporary scholarly concepts of Traditional Chinese Medicine (TCM), Chiding disease was described as an acute symptomatic disease with inevitable serious conditions of convulsion-like or mania-like behavior, and as a refractory ailment. It was also considered as an alias of Hongsiding (namely the acute lymphangitis) in few ancient books. Matixiang, a Chinese herb for treatment of Chiding, was regarded as a nonsuch drug by Da he wai ke, and was considered as a take-then-cure drug by Chuang yang jing yan quan shu (Compendium for Experience in Sores Diseases).

In this study, a preparative chromatography method named preparative argentation reversed-phase high performance liquid chromatography (Ag-RP-HPLC) was developed by adding silver ion to the mobile phase of preparative HPLC. Firstly, an analytical Ag-RP-HPLC method was developed, and the effects of silver content and acid content in the mobile phase on the column efficiency were studied. Based on the method of linear amplification, a preparative Ag-RP-HPLC technique with optimized separation conditions was developed.
