We present a comprehensive account of our efforts directed towards the synthesis of sacubitril, a neprilysin inhibitor used in combination with valsartan and marketed as Entresto™. Our initial approach to the formal synthesis of sacubitril employed a chiral pool strategy, utilizing (S)-pyroglutamic acid as a key building block and Cu(I)-mediated Csp2-Csp3 cross-coupling as a key transformation. Further investigations led to the development of chiral amine transfer (CAT) reagents-based stereoselective synthesis.

A novel platform for the skeletal editing of single C-C bonds via a single-carbon insertion has been developed using diazirines. This strategy involves the photogeneration of arylchlorocarbenes as carbynoid species that undergo site-selective carbene insertion into tertiary C-H bonds and a subsequent Wagner-Meerwein rearrangement promoted by a silver salt. Our skeletal editing strategy based on a formal selective carbyne C-C bond insertion has been demonstrated in six core-to-core conversions, including linear and cyclic benzylic substrates, alkanes and late-stage functionalizations.

Purpose: In image-guided surgery for breast cancer, the representation of the breast deformation between planning and surgery plays a key role. The breast deforms significantly and behaves as a fluid with some constraints. Concretely, the deep fat layer in the breast deforms fluidly due to its incomplete fixation to the chest wall, while the anchoring structures by fascia avoid excessive deformation.

We investigate resonant third-harmonic generation in near-zero index thin films driven out-of-equilibrium by intense optical excitation. Adopting the Landau weak coupling formalism to incorporate electron-electron and electron-phonon scattering processes, we derive a novel set of hydrodynamic equations accounting for collision-driven nonlinear dynamics in sodium. By perturbatively solving hydrodynamic equations, we model third-harmonic generation by a thin sodium film, finding that such a nonlinear process is resonant at the near-zero index resonance of the third-harmonic signal.

We develop a fully vectorial and non-paraxial formalism to describe spontaneous parametric down-conversion in nonlinear thin films. The formalism is capable of treating slabs with a sub-wavelength thickness, describe the associated Fabry-Pérot effects, and even treat absorptive nonlinear materials. With this formalism, we perform an in-depth study of the dynamics of entangled photon-pair generation in nonlinear thin films, to provide a needed theoretical understanding for such systems that have recently attracted much experimental attention as sources of photon pairs.
