Individuals with Li-Fraumeni syndrome carry inherited mutations in the p53 tumor suppressor gene and are predisposed to tumor development. To examine the mechanistic nature of these p53 missense mutations, we generated mice harboring a G-to-A substitution at nucleotide 515 of p53 (p53+/515A) corresponding to the p53R175H hot spot mutation in human cancers. Although p53+/515A mice display a similar tumor spectrum and survival curve as p53+/- mice, tumors from p53+/515A mice metastasized with high frequency. Correspondingly, the embryonic fibroblasts from the p53515A/515A mutant mice displayed enhanced cell proliferation, DNA synthesis, and transformation potential. The disruption of p63 and p73 in p53-/- cells increased transformation capacity and reinitiated DNA synthesis to levels observed in p53515A/515A cells. Additionally, p63 and p73 were functionally inactivated in p53515A cells. These results provide in vivo validation for the gain-of-function properties of certain p53 missense mutations and suggest a mechanistic basis for these phenotypes.

As the demand for high-power-density microelectronics rises, overheating becomes the bottleneck that limits device performance. In particular, the heterogeneous integration architecture can magnify the importance of heat dissipation and necessitate electrical insulation between critical junctions to prevent dielectric breakdown. Consequently, there is an urgent need for thermal interface materials (TIMs) with high thermal conductivity and electrical insulation to address this challenge.

In inertial confinement fusion experiments, hot spot mix caused by hydrodynamic instabilities is a critical performance limitation. Currently, multi-channel Ross filter pair imaging is used to quantitatively diagnose the mix mass of cryogenic hot spots driven by 100 kJ energy, but this method brings significant uncertainty. To measure the level of mix more accurately, we have developed a two-temperature model to modify the fitted bremsstrahlung spectra based on the characteristics of cryogenic implosion hot spots.

High-burden cholera outbreaks, spreading beyond the traditional cholera-endemic countries, have been reported since 2021 in the WHO African region. Member states in the region have committed to the global goal of cholera elimination by 2030. To track progress towards this goal, WHO-African countries adopted a regional cholera prevention and control framework in 2018.

Background: The main technique for identification of free flap perforator vessels is Doppler sonography, which is not always accurate, user dependent and affected by the patient's body habitus.
Methods: Adult patients undergoing head and neck resection and free flap reconstruction at two academic institutions were enrolled. Doppler sonography was used to identify perforators, and were marked using a skin marker.

Surface plasmons offer a promising avenue in the pursuit of swift and localized manipulation of magnetism for advanced magnetic storage and information processing technology. However, observing and understanding spatiotemporal interactions between surface plasmons and spins remains challenging, hindering optimal optical control of magnetism. Here, we demonstrate the spatiotemporal observation of patterned ultrafast demagnetization dynamics in permalloy mediated by propagating surface plasmon polaritons with sub-picosecond time- and sub-μm spatial- scales by employing Lorentz ultrafast electron microscopy combined with excitation through transient optical gratings.
