Background: The presence of activated macrophages (Mphi) is an early and consistent marker of the inflammatory nature of atherosclerotic disease. Dextran-coated superparamagnetic iron oxide particles (SPIO) are avidly endocytosed. These particles have a strong effect on magnetic resonance signal and have been proposed as a non-invasive probe for the presence of early non-occlusive atherosclerotic disease. We describe the extent to which endogenous and exogenous factors regulate Mphi uptake of SPIO particles.

Methods And Results: Cultured murine Mphi-like cells (J744A.1) incubated with SPIO (0, 11.2, 112.0 and 1120 microg Fe/ml) demonstrated significantly reduced SPIO uptake when pretreated with lovastatin to 61% (P < 0.001) and 43% (P = 0.02) of control at 1.0 microM and 17.5 microM lovastatin respectively. Interferon-gamma (IFN-gamma, 1000 U/ml) increased SPIO uptake to 163% of control, P < 0.05. Interleukin-4 (IL-4, 40 ng/ml) also increased uptake (178% of control, P < 0.04). In cells incubated with SPIO in the absence of serum proteins, SPIO uptake fell to 57% of control (P < 0.001).

Conclusions: Uptake of SPIO by activated Mphi is regulated by endogenous cytokines and serum components as well and exogenous lovastatin. Thus, MRI signal changes after SPIO administration may reflect Mphi phagocytic capacity as well as Mphi presence.

Background: Focal nodular hyperplasia (FNH)-like lesions are hyperplastic formations in patients with micronodular cirrhosis and a history of alcohol abuse. Although pathologically similar to hepatocellular carcinoma (HCC) lesions, they are benign. As such, it is important to develop methods to distinguish between FNH-like lesions and HCC.

Hypoxia, a phenomenon that occurs when the oxygen level in tissues is lower than average, is commonly observed in human solid tumors. For oncological treatment, the hypoxic environment often results in radioresistance and chemoresistance. In this study, a new multifunctional oxygen carrier, carboxymethyl hexanoyl chitosan (CHC) nanodroplets decorated with perfluorohexane (PFH) and superparamagnetic iron oxide (SPIO) nanodroplets (SPIO@PFH-CHC), was developed and investigated.

Geochemical speciation and activation risks of Cd, Ni, and Zn in soils with naturally high background in karst regions of southwestern China.

Agricultural soils in karst regions present a remarkable paradox where high geochemical background levels of heavy metals correspond with unexpectedly low crop uptake, challenging traditional risk assessment frameworks and limiting agricultural development. To decode this paradox, we investigated the geochemical speciation of cadmium (Cd), nickel (Ni), and zinc (Zn) in soil-rice systems in southwestern China, which collectively constitute the world's largest continuous karst region and represent diverse soil weathering stages. We employed three chemical extraction methods that revealed reactive pools ranking as Cd (58.

A DNA origami-based enzymatic cascade nanoreactor for chemodynamic cancer therapy and activation of antitumor immunity.

Chemodynamic therapy (CDT) is a promising and potent therapeutic strategy for the treatment of cancer. We developed a DNA origami-based enzymatic cascade nanoreactor (DOECN) containing spatially well-organized Au nanoparticles and ferric oxide (FeO) nanoclusters for targeted delivery and inhibition of tumor cell growth. The DOECN can synergistically promote the generation of hydrogen peroxide (HO), consumption of glutathione, and creation of an acidic environment, thereby amplifying the Fenton-type reaction and producing abundant reactive oxygen species, such as hydroxyl radicals (•OH), for augmenting the CDT outcome.

Background: Superparamagnetic iron oxide nanoparticles (SPIO) are emerging as a viable alternative to technetium and blue dye. Our study was designed to evaluate the correlation between SPIO dose, injection site, and timing with sentinel lymph node (SLN) detection and iron content in retrieved SLNs.

Methods: This study combined individual patient data from three Dutch and five Swedish studies.
