Stereoisomeric cis and trans substrate analogues for Pin1 were designed and synthesized. The central phosphoSer-Pro core of the Pin1 substrate was replaced by cis and trans amide isosteres in Ac-Phe-Phe-pSer-Psi[(Z and E)CH=C]-Pro-Arg-NH(2), 1 and 2, peptidomimetics. They were synthesized on solid phase in 17% yield for the cis analogue 1, and 16% yield for the trans analogue 2. A second trans amide isostere with a C-terminal N-methylamide 3 was synthesized in 7% yield. The protease-coupled Pin1 assay showed that all three compounds inhibited the Pin1 peptidyl-prolyl isomerase (PPIase) enzymatic activity. The cis isostere 1 was 23 times more potent (K(i) = 1.74 +/- 0.08 muM) than its trans counterpart 2 (K(i) = 40 +/- 2 muM) in competitive inhibition of Pin1. These results suggest that the catalytic site of Pin1 binds cis substrates more tightly in aqueous solution. Antiproliferative activity toward the A2780 human ovarian cancer cell line by the cis and trans analogues correlates with Pin1 inhibition results.

This study aims to enhance our understanding of the temporal and spatial processes scales governing the evolutionary diversification of Neotropical birds with Trans- and Cis-Andean populations of the species from South and Central America. Through a multilocus analysis of the mitochondrial (CytB and ND2) and nuclear genes (I7BF, I5BF, and G3PDH) of 41 samples representing six subspecies, we describe the existing molecular lineages of , and estimate their demographic dynamics. We used Ecological Niche Modeling (ENM) with six different algorithms to predict the potential distribution of in both present-day and past scenarios, examining the overlap climatic niche between Cis- and Trans-Andean lineages.

Solvatochromism and cis-trans isomerism in azobenzene-4-sulfonyl chloride.

Solvatochromism exhibited by azobenzene-4-sulfonyl chloride (here abbreviated as Azo-SCl) has been investigated in a series of non-polar, polar-aprotic and polar-protic solvents. The UV-vis spectra of Azo-SCl exhibit two long-wavelength bands, observed at 321-330 nm (band-I) and 435-461 nm (band-II), which are ascribed to the π*-π (S ← S) and π*-n (S ← S) transitions, respectively. The shorter wavelength band indicates a reversal in solvatochromism, from negative to positive solvatochromism, for a solvent with a dielectric constant of 32.
