Published: November 2004

A new electron cryomicroscopic reconstruction of an actin-scruin bundle from Limulus sperm reveals details about the enormous structural plasticity within F-actin. The twist and tilt of the actin subunits show very large deviations from ideal F-actin, providing clues about actin dynamics.

Centro de Estudios Farmacológicos y Botánicos (CEFYBO), Facultad de Medicina-Universidad de Buenos Aires (UBA/CONICET), Buenos Aires, Argentina.

Background: Endocannabinoids like anandamide (AEA), among other lipids, are recognized signaling molecules that participate in reproductive events.

Objectives: Our aims were to characterize orphan G protein-coupled receptor (GPR55) presence; investigate GPR55 activation by AEA and determine GPR55 role in the bovine sperm function.

Materials And Methods: GPR55 presence was assessed by immunocytochemistry.

Reorganization of the flagellum scaffolding induces a sperm standstill during fertilization.

Mammalian sperm delve into the female reproductive tract to fertilize the female gamete. The available information about how sperm regulate their motility during the final journey to the fertilization site is extremely limited. In this work, we investigated the structural and functional changes in the sperm flagellum after acrosomal exocytosis (AE) and during the interaction with the eggs.

Isthmus is the region of the oviduct considered a reservoir for spermatozoa, where they are retained and released synchronously with ovulation. Integrins mediate this interaction, and it is suggested that they regulate the viability and capacitation of spermatozoa. Spermatozoa retained in the oviductal epithelial cells show specific characteristics: normal morphology, intact acrosome and plasma membrane, no DNA fragmentation, and low levels of intracellular Ca2+, and protein phosphorylation at Tyr.

Sea urchin eggs are covered with layers of extracellular matrix, namely, the vitelline layer (VL) and jelly coat (JC). It has been shown that sea urchin eggs' JC components serve as chemoattractants or ligands for the receptor on the fertilizing sperm to promote the acrosome reaction. Moreover, the egg's VL provides receptors for conspecific sperm to bind, and, to date, at least two sperm receptors have been identified on the surface of sea urchin eggs.
