Headless or unucleated spermatozoa were evaluated in semen specimens of 52 fertile men in order to investigate the frequency and the percentage of this morphological abnormality and to estimate the probable correlation of the abnormality with other semen parameters, since there are no available data. The only sperm parameter which correlated with the number of headless spermatozoa was the number of micro round-headed spermatozoa (r= + 0.4432, P<0.001). It is concluded that the high frequency of headless or unucleated spermatozoa in the semen specimens of fertile men, albeit in low percentage, does not seem to have any biological significance. Moreover headless and round-headed spermatozoa could be the outcome of the same pathogenetic mechanism.

Purpose: Polyamine modulating factor 1 binding protein (PMFBP1) acts as a scaffold protein for the maintenance of sperm structure. The aim of this study was further to identify the new role and molecular mechanism of PMFBP1 during mouse spermatogenesis.
Methods And Results: We identified a profile of proteins interacting with PMFBP1 by immunoprecipitation combined with mass spectrometry and demonstrated that class I histone deacetylases, particularly HDAC3 and chaperonin-containing TCP1 subunit 3 (CCT3), were potential interaction partners of PMFBP1 based on network analysis of protein-protein interactions and co-immunoprecipitation.

Acephalic spermatozoa syndrome (ASS) is a rare and severe type of teratozoospermia characterized by the predominance of headless spermatozoa in the ejaculate. However, knowledge about the causative genes associated with ASS in humans is limited. Loss-of-function of SPATA20 has been suggested to result in the separation of the sperm head and flagellum in mice, whereas there have been no cases reporting SPATA20 variants leading to human male infertility.
