Injury to the central nervous system (CNS) and the resulting neuronal loss contribute significantly to morbidity and mortality in human and domestic animal populations. Most insults induce inflammation and the expression of cytokines. The specific roles of these proteins in neurological damage and repair are not completely understood. However, members of the interleukin-1 (IL-1) family have clear therapeutic potential: the IL-1 agonists, IL-1 alpha and IL-1 beta, are induced by CNS injury, and central injection of IL-1 increases, whilst peripheral or central administration of the IL-1 antagonist, IL-1ra, reduces the extent of the damage by more than 50%. The mechanism of action of these cytokines is the subject of intense research. In this review, we summarise approaches that are being used to investigate neuronal cell death, and the contribution of inflammation and cytokines, in particular the IL-1 family, to neurodegenerative disorders.

The complex set of interactions between the immune system and metabolism, known as immunometabolism, has emerged as a critical regulator of disease outcomes in the central nervous system. Numerous studies have linked metabolic disturbances to impaired immune responses in brain aging, neurodegenerative disorders, and brain injury. In this review, we will discuss how disruptions in brain immunometabolism balance contribute to the pathophysiology of brain dysfunction.

Neurons in the central nervous system (CNS) lose regenerative potential with maturity, leading to minimal corticospinal tract (CST) axon regrowth after spinal cord injury (SCI). In young rodents, knockdown of PTEN, which antagonises PI3K signalling by hydrolysing PIP3, promotes axon regeneration following SCI. However, this effect diminishes in adults, potentially due to lower PI3K activation leading to reduced PIP3.

In clinics, physical injuries to the spinal cord cause a temporary motor areflexia below lesion, known as spinal shock. This topic is still underexplored due to the lack of preclinical spinal cord injury (SCI) models that do not use anesthesia, which would affect spinal excitability. Our innovative design considered a custom-made micro impactor that provides localized and calibrated strikes to the ventral surface of the thoracic spinal cord of the entire CNS isolated from neonatal rats.
