Fructose-1,6-bisphosphate (FBP) is a glucose metabolism intermediate that shows a neuroprotective action in animal models of ischemia and other injuries. The intracellular mechanism of FBP on neuroprotection has not been previously defined. Here, we examined whether FBP has a neuroprotective effect against excitotoxicity, and whether it affects the production of reactive oxygen species (ROS), which are involved in the MAPK pathway in cortical neurons. FBP prevented neuronal death in a dose-dependent manner following 24 h of treatment with the excitotoxin, NMDA. After 8 h of NMDA treatment, we observed FBP-induced inhibition of the production of intracellular ROS, and at the earlier time FBP suppressed NMDA-induced p-p38 and p-ERK expression. In addition, MAPK inhibitors reduced NMDA-induced excitotoxicity and also ROS production. Taken together, our results suggest that the neuroprotective effects of FBP could be explained by down-regulation of free radical production through the p38MAPK/ERK pathway.

The hypoxia microenvironment post-myocardial infarction (MI) critically disturbs cellular metabolism and inflammation response, leading to scarce bioenergy supplying, prolonged inflammatory phase and high risk of cardiac fibrosis during cardiac restoration. Herein, an injectable hydrogel is prepared by Schiff base reaction between fructose-1,6-bisphosphate (FBP)-grafted carboxymethyl chitosan (CF) and oxidized dextran (OD), followed by loading fucoidan-coated baicalin (BA)-encapsulated zein nanoparticles (BFZ NPs), in which immunoregulatory and metabolism improving functions are integrally included. The grafted FBP serves to enhance glycolysis and provide more bioenergy for cardiomyocytes survival under hypoxia microenvironment, and elevating cellular antioxidant capacity pentose phosphate pathway.

Deep learning image reconstruction (DLIR) has shown potential to enhance computed tomography (CT) image quality, but its impact on tumor visibility and adoption among radiologists with varying experience levels remains unclear. This study compared the performance of two deep learning-based image reconstruction methods, DLIR and Pixelshine, an adaptive statistical iterative reconstruction-volume (ASIR-V) method, and filtered back projection (FBP) across 33 contrast-enhanced CT staging examinations, evaluated by 20-24 radiologists. The signal-to-noise ratio (SNR) and contrast-to-noise ratio (CNR) were measured for tumor and surrounding organ tissues across DLIR (Low, Medium, High), Pixelshine (Soft, Ultrasoft), ASIR-V (30-100%), and FBP.
