In this study, a novel antibody-drug conjugate (ADC) consisting of Rituximab and Chlorambucil (Rituximab-CMB) was synthesized. The average number of drug molecules attached per Rituximab molecule was determined using MALDI-TOF mass spectrometry, revealing a range of 4-6 drug molecules per antibody. To further improve the therapeutic potential of the ADC, it was radiolabeled with the therapeutic radionuclide Lu via a DOTA chelator, achieving a final radiochemical purity of over 95%.

Background: Biological medications have played a significant role in maintenance therapy for Crohn's disease (CD), but some cases become refractory to these agents. Methotrexate (MTX) appears to be a cost-effective and readily available drug for enhancing the effectiveness of maintenance therapy when used in combination with anti-tumor necrosis factor (anti-TNF) therapy in such cases. Magnetic microrobots are significant platforms for targeted drug delivery, among which sperm-inspired types have attracted much attention due to their flexible undulation. However, mass production of sperm-like soft magnetic microrobots with high-speed propulsion is still challenging due to the need of more reasonable structure design and facile fabrication. Herein, a novel strategy is proposed for large-scale preparation of microalgae-based soft microrobots with a fully magnetic head-to-tail structure, called AlgaeSperm with robust propulsion and chemo-photothermal performance.
