Article Abstract

In haem-regulated phosphodiesterase (PDE) from Escherichia coli (Ec DOS), haem is bound to the PAS domain, and the redox state of the haem iron regulates catalysis by the PDE domain. We generated mutants of Asp40, which forms a hydrogen bond with His77 (a proximal haem axial ligand) via two water molecules, and a salt bridge with Arg85 at the protein surface. The redox potential of haem was markedly increased from 67 mV vs. the standard hydrogen electrode in the wild-type enzyme to 95 mV and 114 mV in the Ala and Asn mutants, respectively. Additionally, the auto-oxidation rate of Ec DOS PAS was significantly increased from 0.0053 to 0.051 and 0.033 min(-1), respectively. Interestingly, the catalytic activities of the Asp40 mutants were abolished completely. Thus, Asp40 appears to play a critical role in the electronic structure of the haem iron and redox-dependent catalytic control of the PDE domain. In this report, we discuss the mechanism of catalytic control of Ec DOS, based on the physico-chemical characteristics of the Asp40 mutants.

Tuning Fork Scanning Electrochemical Cell Microscopy for Resolving Morphological and Redox Properties of Single Ag Nanowires.

We report a Tuning Fork Scanning Electrochemical Cell Microscopy (TF-SECCM) technique for providing morphological and electrochemical information on single redox-active entities. This new operation configuration of SECCM utilizes an electrolyte-filled nanopipette tip mounted onto a tuning fork force sensor to obtain a precise tip-sample distance control and surface morphological mapping capabilities. Redox activities of regions of interest (ROIs) can be investigated by scanning electrode potential by moving the nanopipette to any target regions while maintaining the constant force engagement of the tip with the sample.

Developing new photocatalysts for the selective oxidation of thioethers to high-value-added sulfoxides under low-oxygen mild conditions is a promising but challenging strategy. Here, a new polyoxometalate-based metal-organic framework (POMOF), , was successfully synthesized, wherein continuous π···π stacking interactions and direct coordination bonds not only strengthen the framework's stability but also accelerate electron transfer. A series of experiments and theoretical studies, including control experiments, kinetic studies, electrochemical spectroscopic analyses, and electron paramagnetic resonance, revealed the synergistic catalytic effect among Co(II) metal centers, BWO, and the photosensitizer TPT.

Hybrid polyionic complexes (HPICs) are colloidal structures with a charged core rich in metal ions and a neutral hydrophilic corona. Their properties, whether as reservoirs or catalysts, depend on the accessibility and environment of the metal ions. This study demonstrates that modifying the coordination sphere of these ions can tune the properties of HPICs by altering the composition of the complexing block or varying formulation conditions.
