Methods of obtaining (from cadaveric human brain) the highly-active extracts to be used later as raw-materials for thromboplastin manufacturing, which is applied to determine the prothrombin (thromboplastin) time of human plasma or blood, are described in the paper. The study resulted in defining the optimal requirements to choosing the raw-materials, storing regime, centrifuging and extraction. The thus elaborated technology of soluble thromboplastin manufacturing ensures the production of a reagent with a high sensitivity to a changing level of factors VII and X and to a reducing activity of the prothrombin complex, which is achieved through the impact of indirect anticoagulants.

In 2010, the reporting of thrombotic adverse events for one subcutaneous and certain intravenous immunoglobulins (IGs) raised some concerns. In Europe, regulatory bodies rapidly revised compendial specifications for therapeutic IGs to ensure they do not exhibit thrombogenic (procoagulant) activity (PCA). At the global level, a working group (GWG) was launched with the aim of assessing PCA measurement methods and limits, considering results obtained by human IG manufacturers during in-process controls.

Introduction: An investigation of the suitability of reagents for measuring FVIII products in a one-stage clotting assay (OSA) showed variations in their FVIII activity (FVIII:C). Most studies have focused on the activated partial thromboplastin time (APTT) reagent rather than FVIII-deficient plasma (F8DP), even though the APTT-based OSA is comprised of APTT reagents and factor-deficient plasma.
Aim: A single-centre study was conducted to clarify variations in measurements of FVIII products in an OSA using a total of 12 reagent combinations, including four APTT reagents and three types of F8DP.
