Ipsilateral coordination deficits and central processing requirements associated with coordination as a function of aging.

Young and elderly participants performed concurrent ipsilateral hand-foot movements either isodirectionally or nonisodirectionally. We determined performance by measuring the maximal cycling frequency at which the coordination pattern could be performed successfully (CF(max)). We also determined attentional costs by means of a dual-task paradigm. Findings revealed that CF(max) was significantly lower in the elderly than in the young participants for the nonisodirectional mode, whereas we observed no differences for the isodirectional mode. Under dual-task conditions, coordination deteriorated in the elderly group only. However, when we equated levels of task difficulty, differences between the groups disappeared. Furthermore, attentional costs did not differ between isodirectional and nonisodirectional movements. This indicates that age-related coordination deficits were not primarily evoked by reduced attentional resources or control in elderly persons.

The quantitative evaluation of motor function in experimental stroke models is essential for the preclinical assessment of new therapeutic strategies that can be transferred to clinical research; however, conventional assessment tests are hampered by the evaluator's subjectivity. We present an artificial intelligence-based system for the automatic, accurate, and objective analysis of target parameters evaluated by the ledged beam walking test, which offers higher sensitivity than the current methodology based on manual and visual counting. This system employs a residual deep network model, trained with DeepLabCut (DLC) to extract target paretic hindlimb coordinates, which are categorized to provide a ratio measurement of the animal's neurological deficit.

Spontaneous cleavage reactions normally occur in vivo on amino acid peptide backbones, leading to fragmentation products that can have different physiological roles and toxicity, particularly when the substrate of the hydrolytic processes are neuronal peptides and proteins highly related to neurodegeneration. We report a hydrolytic study performed with the HPLC-MS technique at different temperatures (4 °C and 37 °C) on peptide fragments of different neuronal proteins (amyloid-β, tau, and α-synuclein) in physiological conditions in the presence of Cu and Zn ions, two metal ions found at millimolar concentrations in amyloid plaques. The coordination of these metal ions with these peptides significantly protects their backbones toward hydrolytic degradation, preserving the entire sequences over two weeks in solution, while the free peptides in the same buffer are fully fragmented after the same or even shorter incubation period.
