Micropatterns of spores displaying heterologous proteins. | LitMetric

Micropatterns of spores displaying heterologous proteins.

J Am Chem Soc

Department of Chemical and Biomolecular Engineering, KAIST, Daejeon 305-701, Korea.

Published: September 2004

Micropatterns of Bacillus thuringiensis spores were generated by a combination of surface chemistry, microcontact printing, and spore surface display technique. The outermost layers of B. thuringiensis spores were engineered to present enhanced green fluorescent protein (EGFP), and the biospecific interaction between biotin and streptavidin was utilized to spatially direct the EGFP-presenting spores onto the micropatterned surfaces. The viability of the micropatterned spores was confirmed by the pattern generation of vegetative cells after the germination.

The occurrence of mosquito-borne diseases is increasing, and their geographical range is expanding due to climate change. New control measures are urgently needed to combat these debilitating and, in some cases, fatal diseases. Bacteria of the genus Bacillus are of interest due to the production of bioactive compounds, including those useful for insect control.

For over 100 years, (Bt) has been used as an agricultural biopesticide to control pests caused by insect species in the orders of Lepidoptera, Diptera and Coleoptera. Under nutrient starvation, Bt cells differentiate into spores and associated toxin crystals that can adopt biofilm-like aggregates. We reveal that such Bt spore/toxin biofilms are embedded in a fibrous extrasporal matrix (ESM), and using cryoID, we resolved the structure and molecular identity of an uncharacterized type of pili, referred to here as Fibrillar ENdospore Appendages or 'F-ENA'.

Department of Entomology, Faculty of Agriculture, Kasetsart University, Bangkok, 10900, Thailand.

Organophosphates, carbamates and synthetic pyrethroids are commonly used in Thailand to control mosquito vectors; however, long-term use of insecticides in vector control has led to the rapid development of insecticide resistance. In this study, we screened strains as biological control agents for potential toxins against mosquito larvae as an additional control tool. Preliminary bioassays conducted on 434 strains demonstrated that 41 strains (11.

State Key Laboratory of Ecological Pest Control for Fujian and Taiwan Crops & Key Laboratory of Biopesticide and Chemical Biology of Ministry of Education, College of Life Science and College of Plant Protection, Fujian Agriculture and Forestry University, Fuzhou 350002, Fujian, China. Electronic address:

Bacillus thuringiensis (Bt) is affected by ultraviolet radiation and bacterial sedimentation in pest control applications, leading to low pesticide utilization and a short duration of control. To improve Bt stability in these applications and prolong the duration of biological control, Bt LLP29 was first encapsulated using double emulsion technology, resulting in the formation of W/O/W double emulsion microcapsules with sodium lignosulfonate and chitosan as wall materials. The morphological structure and functionality of microcapsules were then systematically investigated.
