We are proposing to evaluate whether a complementary approach based on cycles of oxygen-ozone autohemotherapy (O3-AHT) already performed in millions of patients, can abate the chronic oxidative stress and improve the quality of life of serious hemoglobinopathic patients. Although a preliminary study has yielded encouraging results, it appears appropriate to perform a controlled, randomized and possibly multicentre clinical trial. The long use of this approach in other pathologies has proved to be very useful and it is hoped that scepticism will not prevail over scientific rationale. Ozone, as any other drug, has an intrinsic toxicity that, in the proposed application, is fully tamed by the blood antioxidant system.

A computational study of X-H···Y binary hydrogen-bonded complexes was undertaken to examine the red- and blue-shifting behavior of three model X-H proton donors interacting with a series of Lewis bases: Y = NH, NCLi, NCH, NCF, CH, BF, CO, N and Ne. Two of these proton donors, FArH and FCH, have blue-shifting tendencies, while the third, FH, has red-shifting tendencies. A perturbation theory model for frequency shifts that was derived many years ago was employed to partition the predicted frequency shift into the sum of two components, one dependent on the second derivative of the interaction energy with respect to X-H displacement and the other dependent on the X-H bond length change in the binary complex.

Proton-pumping rhodopsins, which consist of seven transmembrane helices and have a retinal chromophore bound to a lysine side chain through a Schiff base linkage, offer valuable insights for developing unidirectional ion transporters. Despite identical overall structures and membrane topologies of outward and inward proton-pumping rhodopsins, these proteins transport protons in opposing directions, suggesting a rational mechanism that enables protons to move in different directions within similar protein structures. In the present study, we clarified the chromophore structures in early intermediates of inward and outward proton-pumping rhodopsins.

Clinical trials and cell culture experiments have identified resistance associated mutations (RAMs) in the vicinity of the hydrophobic CA pocket targeted by LEN. The M66I substitution conferred by far the highest level of resistance to the inhibitor compared to other RAMs.

The fusion assembly strategy of supramolecular chemistry combined with dynamic covalent chemistry has provided novel insights into the design of precision nutrition and intelligent drug delivery carriers. This work involved the development of a supramolecular self-assembly originating from entropy- and enthalpy-driven dynamic covalent bonding on Schiff bases between egg white-derived peptide Gln-Ile-Gly-Leu-Phe (QIGLF) and glutaraldehyde (GA), denoted QIGLF-GA. The assembly exhibited outstanding assembly characteristics and multiwavelength autofluorescence properties.
