Objectives: Polymorphisms of transporters associated with antigen-processing (TAP) genes might influence the susceptibility to schizophrenia by altering the antigen-processing pathway. The aim of this study was to verify the relationship between schizophrenia and the polymorphisms of TAP2 genes.

Method: Two hundred and fifty-seven Korean patients diagnosed with schizophrenia according to DSM-IV and 184 normal controls participated in this study. TAP2 polymorphic residues at positions 379, 565 and 665 were typed using amplification refractory mutation system-polymerase chain reaction single-strand conformation polymorphism.

Results: Distribution of the alleles and genotypes in patients with schizophrenia was not significantly different from those of controls.

Conclusions: This study did not show the association of the TAP2 gene with schizophrenia in the Korean population.

Major histocompatibility complex class I deficiency results from deleterious biallelic variants in TAP1, TAP2, TAPBP, and B2M genes. Only a few patients with variant-curated TAP1 deficiency (TAP1D) have been reported in the literature and the clinical phenotype has been variable with an emphasis on autoimmune and inflammatory complications. We report TAP1D in a Nepalese girl with a severe clinical phenotype with serious viral infections at a very young age.

Identification of TAP2 protein variants resistant to inhibition by the HSV1 ICP47 protein.

Department of Biomedical Informatics and the OSU Comprehensive Cancer Center, The Ohio State University, Columbus, OH.

Herpes Simplex Virus 1 evades the host immune system by expressing a protein, ICP47, that binds to and inhibits the heterodimeric Transporter Associated with Antigen Processing (TAP). We screened a library of 1786 variants in TAP2, one of the components of the TAP heterodimer, and identified 39 variants that were resistant to inhibition by ICP47. Of these 39 variants, five were individually tested, and three (T257I, S274H, and T244R) were confirmed to be significantly resistant to inhibition by ICP47.

Article Synopsis
  • Colorectal cancer (CRC) shows a complicated hereditary pattern, with an emphasis on rare genetic variants that could significantly affect CRC susceptibility, particularly in high-risk populations like those in the Middle East.
  • A study involving 146 high-risk CRC patients and 1395 healthy controls sought to find rare genetic variants, revealing that specific inactivating variants had a strong link to high-risk CRC, notably showing a 60-fold increased risk for carriers.
  • The findings suggest that several new genetic variants may influence CRC risks and highlight the need for further large-scale studies to validate these associations and improve genetic counseling for high-risk individuals and their families.

Article Synopsis
  • Genome-wide association studies (GWASs) have identified genetic regions linked to juvenile dermatomyositis (JDM), but the specific functional genes in these regions are still unclear.
  • This study focused on mA-associated SNPs (mA-SNPs) in relevant cell types to determine their impact on gene expression in relation to JDM using bulk tissue and single-cell analyses.
  • Seven mA-SNPs were associated with JDM, revealing differential expression of multiple genes, particularly in myeloid, T, and B cells of JDM patients, highlighting the complex relationship between mA-SNPs and disease susceptibility.
