Four tripeptides (Z-AA1-2Dpy-AA3-OMe; AA1, AA3 = Gly, Aib) containing a novel amino acid, alpha, alpha-di(2-pyridyl)glycine (2Dpy), were synthesized by the modified Ugi reaction. NMR analysis clearly indicated that the 2Dpy-containing tripeptides except the peptide in which AA1, AA3 = Aib, adopt a unique conformation with two intramolecular hydrogen bonds between 2Dpy-NH and a pyridine nitrogen and between AA3-NH and another pyridine nitrogen. This conformation has so far not been reported. On the other hand, the peptide Z-Aib-2Dpy-Aib-OMe probably adopts a beta-turn structure which is stabilized by two intramolecular hydrogen bonds between 2Dpy-NH and a pyridine nitrogen and between AA3-NH and the C=O of the Z group.

Gaussia Luciferase (GLuc) is a renowned reporter protein that can catalyze the oxidation of coelenterazine (CTZ) and emit a bright light signal. GLuc comprises two consecutive repeats that form the enzyme body and a central putative catalytic cavity. However, deleting the C-terminal repeat only limited reduces the activity (over 30% residual luminescence intensity detectable), despite being a key part of the cavity.

A novel series of azo dyes was successfully synthesized by combining amino benzoic acid and amino phenol on the same molecular framework azo linkage. The structural elucidation of these dyes was carried out using various spectroscopic techniques, including UV-vis, FT-IR, NMR spectroscopy, and HRMS. Surprisingly, the aromatic proton in some dyes exhibited exchangeability in DO, prompting a 2D NMR analysis to confirm this phenomenon.

Using photoswitchable molecules to manipulate supramolecular interactions under light illumination has driven advancements in numerous fields, allowing for the strategic alteration of molecular systems. HBT-DPI was a single-molecule multi-conformational fluorescent material and had unique applications for hydrophobic/hydrophilic mapping on large-scale heterogeneous surfaces. In this paper, the different proton transfer processes and luminescence mechanisms of HBT-DPI in Dichloromethane (DCM, no hydrogen bond (HB) receptor) and N, N-Dimethylformamide (DMF, HB receptor) solvents were systematically studied. Using the quantum chemistry method, the stable structures of HBT-DPI in two solvents were determined based on the Boltzmann distribution.
