The crystal structure of the self-complementary DNA octamer d(GGCATGCC) has been determined to a resolution of 2.38 A and R factor of 17.1%. The structure is an A-DNA octamer duplex with Watson-Crick base pairing along its length. This sequence extends the investigation of octamer duplexes of the type d(GGX(4)CC) which assume P6(1) packing within the crystallographic unit cell. The A-DNA duplex is non-linear with a helix axis curvature of 7 degrees and bending towards the DNA minor groove. In view of the biological importance of protein recognition of some A-DNA sequences the minor-groove geometry and helix-axis curvature for this structure is compared with all others of the type d(GGXATXCC).

Reactive oxygen species (ROS) generate DNA lesions that alter genome integrity. Among those DNA lesions, 7,8-dihydro-8-oxo-2'-deoxyguanosine (8-oxodG) is particularly mutagenic. 8-oxodG efficiently incorporates deoxycytidine monophosphate (dCMP) and deoxyadenosine monophosphate (dAMP) via base pairing mediated by its anti and syn conformations, respectively.

Introduction/background: TOP3B (Topoisomerase III-Beta) is a DNA topoisomerase enzyme essential for managing DNA topology during various cellular processes. TOP3B knockout mice typically develop but have a shorter lifespan; however, the exact role of TOP3B is not fully understood. This study aims to investigate the diversity of TOP3B across various mammalian species, with a particular focus on comparing the naked mole-rat (Heterocephalus glaber), known for its exceptional longevity and genomic stability, and humans (Homo sapiens).

The abuse of chlorpyrifos (CPF), an organophosphorus pesticide, poses significant health risks to humans. Therefore, rapid and accurate detection of residual CPF is crucial to human health due to its high risk in trace amounts. Herein, we developed a simple aptasensor that combines a DNA hydrogel-based self-driven capillary with nicking enzyme-mediated amplification (NEMA), in which the NEMA is triggered through the interaction of the aptamer with CPF, and then amplified to produce a large number of single-stranded DNA that can destroy the three-dimensional structure of the DNA hydrogel.

Safety and ethical issues are the primary concerns for assisted reproductive technology (ART). However, confusion and contamination of samples are common problems in embryo laboratories, preimplantation genetic test (PGT) laboratories, and third-party medical testing laboratories due to large sample numbers and complex procedures. Once these problems occur, they are often difficult to trace, posing risks and ethical challenges to hospital reproductive centers, third-party medical testing laboratories, and patient families.

Gene-targeted therapies are revolutionizing cancer treatment due to their high specificity and low toxicity. Among these, ribozymes hold promise as independent gene therapy agents capable of directly cleaving target mRNAs. The pistol ribozyme, discovered in 2015, stands out for its compact structure and robust cleavage activity, making it a promising candidate for RNA silencing under physiological conditions.
