Published: September 2004

Background: Lipid peroxidation is a constant problem that eukaryotic cells have to face. Glutathione peroxidases (GPx) are among the most effective systems that protect cells from hydroperoxide toxicity. The objective of this study was to evaluate the relationship between GPx and cyclooxygenase 2 (COX-2), implicated in cancer pathogenesis, particularly in colon cancer cells.

Materials And Methods: Phospholipid hydroperoxide glutathione peroxidase (PHGPx or GPx4), which metabolizes peroxidized phospholipids, was cloned in an expression plasmid, transfected in HT29 cl.19A colon carcinoma cells and the effects of PHGPx overexpression were measured on arachidonic acid metabolism by COX-2. Metabolites were studied by HPLC and EIA; COX-2 mRNA levels were analysed rising semi-quantitative PCR.

Results: Prostaglandins (PGE2, PGF2alpha, 6 keto-PGF1alpha) and thromboxane (TXB2) production were increased. COX-2 mRNA levels increased in PHGPx overexpressing cells.

Conclusion: Surprisingly, our data suggest that PHGPx overexpression noticeably increases COX-2 metabolism.

Sodium aescinate (SA), a natural plant extract with various bioactivities, is widely used to treat oedema and inflammation in clinics. However, adverse events, including liver injury, kidney injury, and phlebitis, have been reported in patients with SA in recent years. In this study, we used BALB/c mice and L02 cells to evaluate the role of ferroptosis in SA-induced liver injury.

Iron Knights with Nanosword Induced Ferroptosis in the Battle Against Oral Carcinoma.

Article Synopsis
  • Oral squamous cell carcinoma (OSCC) is particularly sensitive to ferroptosis, but traditional inducers have limitations.
  • This study created ultrasmall AuMn nanoclusters (AMNCs) wrapped in NK cell-derived exosomes (Exo-AMNCs) to enhance targeted treatment for OSCC.
  • The AMNCs produce reactive oxygen species to induce ferroptosis while being more biocompatible, and when combined with exosomes, they show effective imaging and therapeutic potential by regulating specific proteins involved in the disease.

Sanggenon c, a component in Morus alba L, has been proved to possess various biological activities. The aim of this study is to investigate whether sanggenon c can target SLC7A11 and inhibit lung cancer by regulating the ferroptosis mechanism. The levels of antioxidant factor, Fe , and SLC7A11 were measured in the lungs of cancerous mice and human A 549 lung cancer cells.

Ferroptosis exacerbates the clonal deletion of virus-specific exhausted CD8 T cells.

Front Immunol

Institute of Immunological Innovation and Translation, Chongqing Medical University, Chongqing, China.

During chronic infection or tumorigenesis, persistent antigen stimulation contributes to the exhaustion of CD8 T cells. Nevertheless, exhausted CD8 T (T) cells still preserve certain effector function, and maintaining a reservoir of exhausted cells is of vital importance for virus elimination and tumor eradiation. Despite considerable work interrogating the rejuvenation of T cells, mechanisms underpinning the clonal deletion of T cells remain largely unexplored over the past decade.

Cerebral infarction is the main cause of death and long-term disability worldwide. Neuronal degeneration and limp sensory dysfunction are the secondary damages induced by cerebral infarction. This study aimed to investigate the roles of activating transcription factor 4 (ATF4) in cerebral infarction and the underlying molecular mechanisms.
