The polyhedrin (polh) gene of nucleopolyhedroviruses (NPVs) encodes for the matrix protein of the virus occlusion body and is one of the most conserved baculovirus genes. Previous analyses of different NPV genes and polh genes provided conflicting results indicating that the Autographa californica nucleopolyhedrovirus (AcMNPV) is generally a member of the so-called group I NPVs and is most closely related to Rachiplusia ou (Ro) NPV, whereas the AcMNPV polh is more similar to the polh of the group II NPVs. A comparative analysis of the AcMNPV polh and its closest neighbours within group I and group II NPV, the RoMNPV and the Thysanoplusia orichalcea (Thor) NPV, was performed using Hidden Markov Models for detecting recombination. The result provided strong evidence that the AcMNPV polh is a chimerical gene which consists of a mosaic of group I and group II NPV specific sequences.

The occlusion bodies of Autographa californica multiple nucleopolyhedrovirus are proteinaceous formations with significant biotechnological potential owing to their capacity to integrate foreign proteins through fusion with polyhedrin, their primary component. The baculoviral chitinase (CHIA) and cathepsin (V-CATH) enzymes promote terminal insect host liquefaction, which aids viral progeny dissemination. Recombinant Autographa californica nucleopolyhedrovirus (AcMNPV)-derived viruses were previously generated with reprogrammed transcription by replacing the native promoter with the AcMNPV () or core protein () promoter sequences, but of both these -reprogrammed viruses lacked transcription and V-CATH enzymatic activity. Here, we report that dual / promoter reprogramming of the adjacent genes resulted in modulated temporal transcription of both genes without impacting infectious budded virus production.

Early and late gene expressions of baculoviruses have been known to rely on host RNA polymerase II and a virus-encoded RNA polymerase, separately. In this study, we found that Autographa californica multiple nucleopolyhedrovirus (AcMNPV) recombinant bacmids with the individual RNA polymerase subunit genes deleted could support low levels of expression of a reporter gene under the control of the promoter of a typical late gene, , in transfected Sf9 cells. Through multistep subcloning of a genomic library of the virus and transient expression assay analysis, was identified to be the only viral gene that was responsible for activation of late gene expression in the absence of the viral RNA polymerase.

PlxyMNPV_LBIV-11 is an alphabaculovirus strain, isolated from Plutella xylostella larvae. This work characterized this strain at a biological, morphological, and molecular level to evaluate its similarity with other baculoviruses. Its ultrastructure showed a multiple arrangement of nucleocapsids within enveloped virions, all occluded within large cubical polyhedra.
