Bacteriophages that infect the cellulolytic ruminal bacterium Ruminococcus albus AR67.

Published: August 2004

Aim: To isolate bacterial viruses that infect the ruminal cellulolytic bacterium Ruminococcus albus.

Methods: Four phages infecting R. albus AR67 were isolated under anaerobic conditions using the soft-agar overlay technique. The phages were characterized on morphology, solvent stability, nucleic acid type and digestion characteristics. Two phages, phiRa02 and phiRa04 comprised icosahedral virions with linear double-stranded DNA and appeared to belong to the family Podoviridae [corrected] The other two phages are most likely filamentous phages with circular single-stranded DNA of the family Inoviridae.

Significance Of The Study: Viruses of the family Inoviridae [corrected] have not previously been isolated from rumen bacteria. The phages isolated in this study are the first phages shown to infect the cellulolytic bacteria of the rumen. This suggests that the cellulolytic populations of the rumen are subject to lytic events that may impact on the ability of these bacteria to degrade plant fibre and on the nutrition of the animal.

Several mycobacterial species are known to cause human diseases, such as tuberculosis and leprosy. In addition to these pathogenic species, there are also saprophytic representatives, which occasionally cause opportunistic infections. It is well established that numerous mycobacteria produce biofilms containing cellulose, and their genomes frequently harbor genes involved in cellulose degradation, such as celA1.

Article Synopsis
  • Researchers found that a new strain called LZ117 is highly effective at producing enzymes that break down plant biomass and can also help control pests and plant diseases.
  • They used DNA analysis to confirm that LZ117 is a unique species related to others in its group, and characterized its genetic makeup, including genes linked to enzyme production and transport mechanisms.
  • The study showed that genetic manipulation of LZ117 is possible, including overexpressing a transporter gene, which does not hinder enzyme production but highlights the role of a specific gene in regulating enzyme production under glucose conditions.

Article Synopsis
  • * The Parangkusumo sand dunes in Java, Indonesia, represent a coastal ecosystem with extreme conditions and have not previously had PGPR studied, making this research significant.
  • * The study isolated ten rhizobacterial strains from Spinifex littoreus, with several showing beneficial activities like phosphate solubilization and nitrogen fixation, which could help create effective biofertilizers for coastal farming.

Acute hepatopancreatic necrosis disease, caused by strains carrying the A and B toxin genes (VpAHPND), has been causing great economic losses in Asia and America in the shrimp farming industry. Numerous strains are resistant to antibiotics. However, supplementation with probiotic antagonists has become a more desirable treatment alternative.

Anaerobic protists frequently harbour methanogenic archaea, which apparently contribute to the hosts' fermentative metabolism by consuming excess H. However, the ecological properties of endosymbiotic methanogens remain elusive in many cases. Here we investigated the ecology and genome of the endosymbiotic methanogen of the protists in the hindgut of the termite .
