The contributors to this discussion were invited to submit comments, each from a different standpoint, on the paper by John Caldwell and Thomas Schindlmayr that appeared in the preceding issue of the journal. The invitation was issued with the approval of these authors, and the journal is grateful to them for allowing their paper to be used to generate debate on the issues they had raised. The discussion is followed by the authors' response to it.

Article Synopsis
  • This study analyzed the relationship between insomnia and sleep apnea with deployment and combat exposure in active-duty soldiers from 2010 to 2019, involving over 1.2 million soldiers.
  • The data showed that incidents of insomnia increased from 2010 to 2012, then declined, while sleep apnea rates increased until 2016, but both disorders remained common among soldiers.
  • Findings indicated that both deployment and combat exposure significantly raised the risks of developing insomnia and sleep apnea, with soldiers’ sleep issues highlighting the need for ongoing screening and support for better sleep habits.

Direct dorsal root ganglia (DRG) injection in mice for analysis of adeno-associated viral (AAV) gene transfer to peripheral somatosensory neurons.

Background: Delivering optogenetic genes to the peripheral sensory nervous system provides an efficient approach to study and treat neurological disorders and offers the potential to reintroduce sensory feedback to prostheses users and those who have incurred other neuropathies. Adeno-associated viral (AAV) vectors are a common method of gene delivery due to efficiency of gene transfer and minimal toxicity. AAVs are capable of being designed to target specific tissues, with transduction efficacy determined through the combination of serotype and genetic promoter selection, as well as location of vector administration.

Rationale: Behavioral effects of testosterone depend on dose, acute versus sustained formulation, duration of administration, personality, genetics, and endogenous levels of testosterone. There are also considerable differences between effects of endogenous and exogenous testosterone.

Objectives: This study was the secondary behavioral arm of a registered clinical trial designed to determine if testosterone protects against loss of lean body mass and lower-body muscle function induced by a severe energy deficit typical of sustained military operations.

High hit rates from initial ligand-observed NMR screening can make it challenging to prioritize which hits to follow up, especially in cases where there are no available crystal structures of these hits bound to the target proteins or other strategies to provide affinity ranking. Here, we report a reproducible, accurate, and versatile quantitative ligand-observed NMR assay, which can determine values of fragments in the affinity range of low μM to low mM using transverse relaxation rate as the observable parameter. In this study, we examined the theory and proposed a mathematical formulation to obtain values using non-linear regression analysis.
