A new liquid chromatographic-mass spectrometric (LC-MS) method for determining trace concentrations of gamma-hydroxybutyric acid (GHB) in biological samples has been developed. This method utilizes solid-phase extraction for separation, deuterated GHB as an internal standard (IS) and multiple reaction monitoring (MRM) in the negative ion mode to detect the parent and product ions (103 and 57 for GHB, and 109 and 61 for D6-GHB, respectively). The assay produces excellent linearity and reproducibility, with a limit of quantification (LOQ) of about 0.1 microg/ml. The method has been applied for the determination of endogenous GHB in various rat brain regions.

A simple analytical workflow is described for gas chromatographic-mass spectrometric (GC-MS)-based chiral profiling of secondary amino acids (AAs) in biological matrices. The sample preparation is carried out directly in aqueous biological sample extracts and involves in situ heptafluorobutyl chloroformate (HFBCF) derivatization-liquid-liquid microextraction of nonpolar products into hexane phase followed by subsequent formation of the corresponding methylamides from the HFB esters by direct treatment with methylamine reagent solution. The (O, N) HFB-butoxycarbonyl-methylamide AA products (HFBOC-MA) are separated on a Chirasil-L-Val capillary column and quantitatively measured by GC-MS operated in selected ion monitoring (SIM) mode.

Sebum is a biofluid excreted by sebaceous glands in the skin. In recent years sebum has been shown to contain endogenous metabolites diagnostic of disease, with remarkable results for Parkinson's Disease. Given that sebum sampling is facile and non-invasive, its potential for use in clinical biochemistry diagnostic assays should be explored including the parameters for standard operating procedures around collection, transport, and storage.
