In Parkinson's disease and other neurodegenerative diseases, (-)deprenyl, an inhibitor of type B monoamine oxidase (MAO-B), has been proposed to protect or rescue declining neurons. However, clinical trials failed to confirm the neuroprotection, even though in vivo and in vitro studies suggested the possibilities. This paper describes the activities of propargylamine MAO-B inhibitors against apoptosis induced by an endogenous selective dopaminergic neurotoxin, N-methyl(R)salsolinol, in dopaminergic SH-SY5Y cells. A series of propargylamines were shown to suppress the apoptotic cascade; preventing collapse of mitochondrial membrane potential, activation of caspase 3 and fragmentation of nucleosomal DNA. Among propargylamines, (R)-N-propargyl-1-aminoindan (rasagiline) was the most potent at preventing cell death. Rasagiline also prevented opening of permeability transition pore in insolated mitochondria. These results suggest that rasagiline and other propargylamines may regulate the apoptotic machinery in mitochondria and rescue or protect deteriorated neurons in neurodegenerative disorders.

View Article and Find Full Text PDFBackground: Aggregated TDP-43 is found as the main component of pathological inclusions in amyotrophic lateral sclerosis (ALS), limbic-predominant age-related TDP-43 encephalopathy (LATE), in 50% of patients with frontotemporal dementia (FTD), and is present as co-pathology in other neurodegenerative diseases, including Alzheimer's disease (AD). Biofluid or imaging biomarkers of TDP-43 pathology are currently unavailable. Direct detection of TDP-43 aggregates holds promise for unraveling the pathobiology of disease and for enabling precision medicine approach via improved diagnosis, patient stratification and assessment of therapeutic efficacy in clinical trials in AD and related diseases.

Background: In recent decades, epidemiological and experimental studies have looked into the role of pesticides, particularly the herbicide paraquat, in the development of Parkinson's disease. Horseradish tree (Moringa oleifera) is an ethnobotanical plant with lots of therapeutic potential, but there is a dearth of information on the bioactive properties of the seed alkaloid extracts.
Method: This study examined the modulatory effects of various concentrations of an alkaloid extract from the seeds of Horseradish Tree (Moringa oleifera) on the survival rate of flies exposed to paraquat, as well as certain biochemical and molecular markers related to Parkinson's disease in the heads of the flies.

Background: The effect of high consumption of psychoactive substances of codeine (CDE), tramadol (TMD), and Cannabis sativa (CNB) as concoction has been associated with altered brain cognitive and neurochemical functions. However, the understanding of the complex mechanism behind the intake of Cannabis sativa co-administration with tramadol and codeine on both cardiac and brain function, neurotransmitters, purinergic, and antioxidant enzymes activities in the brain and heart of rats remains unreported.
Method: The measure of cognition using morris water maze (MWM) and Y-maze tests, hemodynamic parameters namely systolic blood pressure (SBP) and heart rate (HR), acetylcholinesterase (AChE), butyl-cholinesterase (BCHE), adenosine deaminase (ADA), arginase, catalase (CAT), superoxide dismutase (SOD) enzymes' activities, reduced glutathione (GSH) and malondialdehyde (MDA), nitric oxide (NO) levels, in the brain and heart of CNB, TMD, and CDE exposed rats was done.
