In this paper, we report (i) the in situ localization, and (ii) meal time related and starvation induced changes in preprogalanin mRNA expression in the goldfish brain. The specific brain nuclei that express galanin mRNA are the area ventralis telencephali pars ventralis, nucleus preopticus periventricularis, nucleus lateralis tuberis, and the nucleus recessus lateralis. No changes in preprandial preprogalanin mRNA expression were found in the brain regions studied. No changes in postprandial preprogalanin mRNA expression were found compared to the preprandial levels. However, in unfed fish, a significant increase in preprogalanin mRNA expression was found in the telencephalon (1 and 3 h) and hypothalamus (3 h) after the scheduled feeding time. The postprandial preprogalanin mRNA expression in the telencephalon and hypothalamus of fed fish at 1 and 3 h were significantly lower than the mRNA expression levels in the unfed fish at the same time. Preprogalanin mRNA expression levels remain unaltered during 7 days of starvation. The presence of preprogalanin mRNA in brain nuclei involved in the regulation of food intake and pituitary hormone secretion suggests important physiological roles for galanin in goldfish.

The retrotrapezoid nucleus (RTN) is a hub for respiratory chemoregulation in the mammal brainstem that integrates chemosensory information from peripheral sites and central relays. Chemosensitive neurons of the RTN express specific genetic and molecular determinants, which have been used to identify RTN precise location within the brainstem of rodents and nonhuman primates. Based on a comparative approach, we hypothesized that among mammals, neurons exhibiting the same specific molecular and genetic signature would have the same function.

Long-term hypercapnia is associated with respiratory conditions including obstructive sleep apnea, chronic obstructive pulmonary disease and obesity hypoventilation syndrome. Animal studies have demonstrated an initial (within hours) increase in ventilatory drive followed by a decrease in this response over the long-term (days-weeks) in response hypercapnia. Little is known about whether changes in the central respiratory chemoreflex are involved.

The retrotrapezoid nucleus (RTN) contains CO(2) -responsive neurons that regulate breathing frequency and amplitude. These neurons (RTN-Phox2b neurons) contain the transcription factor Phox2b, vesicular glutamate transporter 2 (VGLUT2) mRNA, and a subset contains preprogalanin mRNA. We wished to determine whether the terminals of RTN-Phox2b neurons contain galanin and VGLUT2 proteins, to identify the specific projections of the galaninergic subset, to test whether RTN-Phox2b neurons contact neurons in the pre-Bötzinger complex, and to identify the ultrastructure of these synapses.

The ventrolateral medulla oblongata (VLM) of the brainstem contains neurochemically heterogeneous neurons that have a critical role in cardiovascular and respiratory regulation. Previous anatomical studies have shown the existence of galanin immunoreactivity in the medulla oblongata, but a detailed characterization is lacking. In this study, we demonstrate three populations of preprogalanin mRNA (PPG)-expressing neurons in the VLM of the adult, male Sprague-Dawley rat: a retrotrapezoid nucleus (RTN) group, a group in the rostral ventral respiratory group (rVRG), and a subpopulation of A1 neurons.
