Two Italian subjects, aged three and sixteen years, presented with early iron overload as shown by increased serum iron indices and hepatic iron concentration. They both carried the Y250X mutation of the TFR2 gene in the homozygous state. We suggest that transferrin receptor-2 is important in maintaining iron balance in the first decades of life.

Purpose: Sepsis is a life-threatening condition associated with acute organ dysfunction. Iron is an essential trace element for multicellular organisms and almost all microorganisms, and its role in sepsis has been increasingly recognized. The aim of this study was to investigate the changes in iron metabolism in caecal ligation and puncture solution (CLP) -induced septic mice and the effects of hepcidin pretreatment on serum inflammatory marker levels and liver iron metabolism in CLP-induced septic mice.

Osteoarthritis (OA) is a prevalent degenerative joint disease affecting over 530 million individuals worldwide. Recent studies suggest a potential link between iron overload, a condition characterised by the excessive accumulation of iron in the body, and the onset of OA. Iron is essential for various biological processes, and any disruption in its homeostasis can trigger significant health effects, including OA.

Article Synopsis
  • - Iron is crucial for various metabolic functions in mammals, with its levels regulated primarily by hepcidin, influenced by proteins like Hfe and TfR2 found in macrophages involved in iron storage and delivery.
  • - Mice lacking Hfe in macrophages show mild iron deficiency and increased production of the iron exporter Ferroportin 1 (Fpn1), affecting their iron metabolism and immune responses, particularly in aged subjects.
  • - The study found that silencing Hfe and TfR2 in macrophages impacted iron levels in serum and liver, with notable differences in iron gene transcription and protein levels between adult and aged mice, highlighting the role of these proteins in regulating iron homeostasis and Erythropoietin

This study provides a comprehensive overview of hereditary hemochromatosis (HH), a genetic condition characterized by iron overload due to excessive iron absorption. It elucidates diverse inheritance patterns and clinical manifestations by exploring mutations in critical genes such as HFE (hemochromatosis), HJV (hemojuvelin), HAMP (hepcidin antimicrobial peptide), TfR2 (transferrin receptor 2), and FP (ferroportin). The significance of early screening, diagnosis, and personalized management strategies based on genetic classification is emphasized, particularly in terms of high-income vs.

Article Synopsis
  • Hereditary hemochromatosis (HH) is a serious iron-storage disorder that can cause significant organ damage, and HH type 3 is linked to mutations in the TFR2 gene, which is rare in Asia.
  • A 69-year-old Japanese woman with joint pain and liver issues was found to have high iron levels and a novel homozygous mutation in her TFR2 gene, identified through genetic testing.
  • After treatment with phlebotomy, her arthritis and liver enzyme levels improved, marking the first documented case of this specific mutation in an HH type 3 patient.
