Our understanding of basic mechanisms of differentiation has evolved rapidly in the last two decades. Spurred by advances in molecular biology and other research technologies, these advances have become of heightened importance with the recent advent of the possibility of engineering different types of stem cells into needed cell and tissue sources. As pediatric surgeons, we have the potential to play a key role in interfacing between the basic science necessary to understand differentiation processes, and its application at the bedside. In this brief article, we outline our in-depth analysis of mechanisms of basic differentiation of pancreatic precursor cells in an effort to better understand ways in which we can engineer a stem cell pool to form mature pancreatic cells.

We immunized three groups of Mojave desert tortoises (Gopherus agassizii): a group immunized twice, a group immunized once, and a group sham-immunized. We used the antigen, ovalbumin (OVA), with Freund's adjuvant to elicit antibody responses similar to those induced by extracellular bacteria. All tortoises have relatively high levels of B1 lymphocytes and natural antibodies (NAbs), and the goal of this study was to quantify B2 lymphocyte activity (antibody production and potential proliferation) that occurs in primary and secondary immunizations against this constitutive, first line of humoral defense.

The vertebrate bauplan is primarily established via the formation of embryonic tissues in a head-to-tail progression. The mechanics of this elongation, which requires the presomitic mesoderm (PSM), remain poorly understood. Here, we find that avian PSM explants can elongate autonomously when physically confined in vitro, producing a pushing force promoting posterior elongation of the embryo.

Keloids are abnormal scars formed due to fibroblast dysfunction and excessively deposited extracellular matrix (ECM). Despite the unclear process leading to the occurrence of keloids, several studies have demonstrated that histamine and its H1 receptor can effectively regulate fibroblast functions, contributing to keloid formation. Chlorpheniramine maleate (CPM) as a first-generation H1 antihistamine has been widely applied in symptomatic treatment of allergic conditions but its effects on keloids are unknown.

The condition in which the insulin secretory ability of pancreatic β-cells decreases in diabetes is extremely important, but there are currently no biomarkers that reflect pancreatic β-cell failure. Therefore, we conducted a search for biomarkers, using pancreatic β-cell-specific 3-Phosphoinositide-dependent protein kinase 1 (PDK1) knockout mice, which develop severe hyperglycemia due to a decrease in pancreatic β-cell mass without insulin resistance. The analysis was performed in young mice when metabolic abnormalities were not yet apparent.
