The enzymes and control of eukaryotic mRNA turnover.

The degradation of eukaryotic mRNAs plays important roles in the modulation of gene expression, quality control of mRNA biogenesis and antiviral defenses. In the past five years, many of the enzymes involved in this process have been identified and mechanisms that modulate their activities have begun to be identified. In this review, we describe the enzymes of mRNA degradation and their properties. We highlight that there are a variety of enzymes with different specificities, suggesting that individual nucleases act on distinct subpopulations of transcripts within the cell. In several cases, translation factors that bind mRNA inhibit these nucleases. In addition, recent work has begun to identify distinct mRNP complexes that recruit the nucleases to transcripts through different mRNA-interacting proteins. These properties and complexes suggest multiple mechanisms by which mRNA degradation could be regulated.

To investigate the regulatory mechanism mediated by hypothalamic OPN5 on seasonal changes in the reproductive activities of domestic geese, 60 Magang ganders in their breeding period were selected for the experiment and evenly divided into an immunization group(OPN5-IM) and a control group. On days 0, 15 and 30, ganders in the immunized group were immunized with OPN5-KLH protein vaccine, and ganders in the control were immunized with the same amount of blank emulsified vaccine. Additionally, 120 female geese were provided to stimulate the reproductive activities of male geese.

The knowledge on the life cycle of flaviviruses is still incomplete, and no direct-acting antivirals against their infections are clinically available. Herein, by screening via a Zika virus (ZIKV) replicon assay, we found that the N-terminus of NS2A exhibited great tolerance to the insertions of different split fluorescent proteins (split-FPs). Furthermore, both ZIKV and dengue virus encoding a split-FP-tagged NS2A propagated efficiently, and the split-FP-tagged ZIKVs had good genetic stability.
