Nonspecific antibodies, which are thought to be nonprotective, have been shown to contribute a substantial proportion of the measured concentration in the standardized immunoglobulin G (IgG) enzyme-linked immunosorbent assay (ELISA) for pneumococcal polysaccharide capsular antibodies. The presence of such antibodies in human immunodeficiency virus (HIV)-infected persons has not been evaluated. The amount of nonspecific antibodies is proportional to the reduction in IgG antibody concentration that occurs with serum absorption with the heterologous polysaccharide 22F. We measured the amount of nonspecific antibodies before and after vaccination with the pneumococcal conjugate vaccine (PCV; n = 33) or the pneumococcal polysaccharide vaccine (PPV; n = 34) in HIV-infected adults with CD4 counts of >/== 200 cells/mm3. Blood was drawn before and 2 months after vaccination. For prevaccination sera, we found a substantial amount of nonspecific antibodies for serotypes 4, 6B, 9V, and 23F (23 to 47% of measured IgG concentration), but not for serotype 14. There tended to be proportionately less nonspecific antibodies in postvaccine sera than prevaccine sera for PCV, but not for PPV. Subjects with a low HIV viral load (

In order to further investigate the mechanism of action of rituximab in humans and its biotransformation, it is essential to obtain high purity rituximab while maintaining its structural and functional integrity. In this study, we proposed and fabricated a small-molecule 4-vinylpyridine-modified high-density membrane material (P4VP@M) using the surface-initiated atom transfer radical polymerization (SI-ATRP) method. This approach can overcome the limitations of conventional enrichment materials, such as long incubation time and low adsorption capacity.

Background: Rheumatic and musculoskeletal diseases, which are prevalent in nearly 25-30% of the Indian population, pose a significant burden on public health. However, the field is often overlooked in undergraduate medical curricula, leading to a shortage of adequately trained healthcare professionals to address these conditions, especially at the primary health care level. Enhancing rheumatology training for undergraduates in India is imperative.

Original Antigenic Sin in CD4+ T Cells.

Immunology

March 2025

School of Mathematics and Statistics, Donghua University, Shanghai, China.

Original antigenic sin (OAS) describes the phenomenon in which prior exposure to an antigen weakens the adaptive antibody response to a subsequent heterologous infection. This phenomenon can diminish the effectiveness of immunity acquired through vaccination or previous infections. We demonstrate that OAS arises because CD4+ T cell proliferation and regulation signals are antigen-nonspecific.

Diagnosis and Treatment of Hypophosphatasia.

Calcif Tissue Int

March 2025

Division of Clinical Genetics, Children's Mercy Kansas City, Kansas City, MO, USA.

Hypophosphatasia (HPP) is a rare inherited metabolic disorder characterized by deficient activity of tissue-nonspecific alkaline phosphatase (TNAP) caused by variants in the ALPL gene. Disease manifestations encompass skeletal hypomineralization with rickets and lung hypoplasia, vitamin B6-dependent seizures, craniosynostosis, and premature loss of deciduous teeth. The clinical presentation can comprise failure to thrive with muscular hypotonia, delayed motor development, and gait disturbances later in childhood.

Background And Aim: The IPB-D2 chicken, a potential disease-resistant local Indonesian line, has been developed to improve poultry health and productivity for small-scale farmers. This study aimed to characterize the disease resistance traits of IPB-D2 chickens by analyzing key immunological parameters, including immunoglobulin Y (IgY) concentration, Newcastle disease (ND) antibody titers, and leukocyte profiles.

Materials And Methods: A total of 100 IPB-D2 chickens were studied.
