Multiple co-crystal structures of an adenine-based series of inhibitors bound to the molecular chaperone Hsp90 have been determined. These structures explain the observed SAR for previously described compounds and new compounds, which possess up to 8-fold improved potency against the isolated enzyme. Anti-tumour cell potency and mechanism of action data is also described for the most potent compounds. These data should enable the design of more potent Hsp90 inhibitors.

Glioblastoma (GB) is one of the most aggressive and treatment-resistant cancers due to its complex tumor microenvironment (TME). We previously showed that GB progression is dependent on the aberrant induction of chaperone-mediated autophagy (CMA) in pericytes (PCs), which promotes TME immunosuppression through the PC secretome. The secretion of extracellular matrix (ECM) proteins with anti-tumor (Lumican) and pro-tumoral (Osteopontin, OPN) properties was shown to be dependent on the regulation of GB-induced CMA in PCs.

The heat shock protein 70 (HSP70) family plays an important role in the growth and development of lettuce and in the defense response to high-temperature stress; however, its bioinformatics analysis in lettuce has been extremely limited. Genome-wide bioinformatics analysis methods such as chromosome location, phylogenetic relationships, gene structure, collinearity analysis, and promoter analysis were performed in the gene family, and the expression patterns in response to high-temperature stress were analyzed.
