Arylalkylamine N-acetyltransferase (AA-NAT) is the key regulatory enzyme in the melatonin biosynthetic pathway. Previous investigations have reported that Aa-nat mRNA in rat is only detected in a sub-population of photoreceptor cells that resemble cones in shape and size. In the present study, we investigated Aa-nat expression in the rat retina by using in situ hybridization and laser capture microdissection combined with the reverse transcription/polymerase chain reaction technique. Our results demonstrate that, contrary to previous reports, Aa-nat transcripts are present not only in the photoreceptor cells, but also in the inner nuclear layer and in the ganglion cell layer. However, the rhythmic expression of Aa-nat mRNA was observed only in photoreceptor cells.

Early-life morphine exposure causes a variety of behavioural and physiological alterations observed later in life. In the present study, we investigated the effects of prenatal and early postnatal morphine on the maturation of the circadian clockwork in the suprachiasmatic nucleus and the liver, and the rhythm in aralkylamine N-acetyltransferase activity in the pineal gland. Our data suggest that the most affected animals were those born to control, untreated mothers and cross-fostered by morphine-exposed dams.

The study was designed to determine the effect of proinflammatory cytokine, interleukin- (IL-) 1β, on melatonin release and expression enzymes essential for this hormone synthesis: arylalkylamine-N-acetyltransferase (AA-NAT) and hydroxyindole-O-methyltransferase (HIOMT) in ovine pineal gland, taking into account the immune status of animals before sacrificing. Knowledge about melatonin synthesis and its potential roles within the retina remains fragmented, especially in mammals where studies have focused on the penultimate enzyme of melatonin synthesis arylalkylamine N-acetyltransferase (AA-NAT), whereas the final enzyme necessary for melatonin production is hydroxyindole-O-methytransferase (HIOMT). We explored multiple parameters of the melatonin synthetic pathway in the cone-rich retina of a diurnal rodent, Arvicanthis ansorgei, cones being previously implicated as probable reservoirs of melatonin production. We analyzed the temporal and spatial expression of Aa-nat mRNA and AA-NAT protein and enzymatic activity of AA-NAT, HIOMT, as well as the melatonin receptor type 1 and melatonin itself.
