Notch signals are important for lymphocyte development but downstream events that follow Notch signaling are not well understood. Here, we report that signaling through Notch modulates the turnover of E2A proteins including E12 and E47, which are basic helix-loop-helix proteins crucial for B and T lymphocyte development. Notch-induced degradation requires phosphorylation of E47 by p42/p44 MAP kinases. Expression of the intracellular domain of Notch1 (N1-IC) enhances the association of E47 with the SCF(Skp2) E3 ubiquitin ligase and ubiquitination of E47, followed by proteasome-mediated degradation. Furthermore, N1-IC induces E2A degradation in B and T cells in the presence of activated MAP kinases. Activation of endogenous Notch receptors by treatment of splenocytes with anti-IgM or anti-CD3 plus anti-CD28 also leads to E2A degradation, which is blocked by the inhibitors of Notch activation or proteasome function. Notch-induced E2A degradation depends on the function of its downstream effector, RBP-Jkappa, probably to activate target genes involved in the ubiquitination of E2A proteins. Thus we propose that Notch regulates lymphocyte differentiation by controlling E2A protein turnover.

We have introduced the floxed allele of Smoothened (Smo) carried by the mouse line Smo into the C57BL/6J strain by serial backcross. Recapitulation of the Smo null phenotype was confirmed by deleting the allele using E2a-cre and intercrossing heterozygous Smo ± mice. No homozygous mutant embryos were identified at E9.

Enhances the resistance of rice to lepidopteran pests by fusing the Cry1Ca and Cry2Aa genes with self-cleavage peptide sequence.

Key Laboratory of Agro-ecological Processes in Subtropical Region, Institute of Subtropical Agriculture, Chinese Academy of Sciences, Changsha, China.

Background: Accumulation of two or more Bacillus thuringiensis (Bt) proteins in plant not only improves the resistance to pests and broadens the resistance spectrum of crops, but also delays the development of pest resistance.

Results: The self-cleavage peptide sequence was used to link two codon-optimized genes, so as to achieve simultaneous accumulation of two low homologous insecticidal proteins in one plant. The rice transformants accumulating Cry1Ca and Cry2Aa proteins were fed to local lepidopteran pests and the larva mortality in 5 days were 100%.

Central Mechanisms of Thermoregulation and Fever in Mammals.

Adv Exp Med Biol

Department of Integrative Physiology, Nagoya University Graduate School of Medicine, Nagoya, Japan.

Thermoregulation is a fundamental homeostatic function in mammals mediated by the central nervous system. The framework of the central circuitry for thermoregulation lies in the hypothalamus and brainstem. The preoptic area (POA) of the hypothalamus integrates cutaneous and central thermosensory information into efferent control signals that regulate excitatory descending pathways through the dorsomedial hypothalamus (DMH) and rostral medullary raphe region (rMR).

Smarcd1 subunit of SWI/SNF chromatin-remodeling complexes collaborates with E2a to promote murine lymphoid specification.

Lymphocyte development from murine hematopoietic stem cells (HSCs) entails a loss of self-renewal capacity and a progressive restriction of developmental potential. Previous research from our laboratory suggests that specialized assemblies of ATP-dependent SWI/SNF chromatin-remodeling complexes play lineage-specific roles during murine hematopoiesis. Here, we demonstrate that the Smarcd1 subunit is essential for specification of lymphoid cell fate from multipotent progenitors.

Early B cell lymphopoiesis depends on E2A, Ebf1, Pax5 and Ikaros family members. In the present study, we used acute protein degradation in mice to identify direct target genes of these transcription factors in pro-B, small pre-B and immature B cells. E2A, Ebf1 and Pax5 predominantly function as transcriptional activators by inducing open chromatin at their target genes, have largely unique functions and are essential for early B cell maintenance.
