Antithrombin and tissue factor pathway inhibitor (TFPI) provide different anticoagulant mechanisms. Having established a potent anticoagulant role of cultured human umbilical vein endothelial cells in vessel-on-a-chip microfluidic models, we now investigated how these cells modulated thrombin generation under stasis through antithrombin and TFPI pathways. We observed that endothelial monolayers in 96 well-plates strongly delayed and suppressed the thrombin generation process induced by tissue factor, regardless of the presence of whole blood, platelet-rich plasma or platelet-free plasma.

Article Synopsis
  • Heparins, derived from animal tissues, have supply limitations and risks like bleeding and thrombocytopenia, prompting research into synthetic alternatives.
  • A new approach synthesized a heparin-like anticoagulant, sulfated octasaccharide (SRO), demonstrating effective anticoagulant activity in mice and humans with less bleeding risk.
  • This synthetic product, named octaparin, offers a promising replacement for traditional animal-sourced heparin.

Article Synopsis
  • A polysaccharide named CZS-0-1 was extracted from the marine green algae Codium fragile using specific chromatography techniques, revealing it to be a sulfated galactoarabinan with an arabinose, galactose, and glucose ratio of 9:2:1 and 21% sulfate content.
  • Structural analysis showed that the backbone consisted mainly of (1 → 3)-β-L-Arap, with branches and sulfate modifications enhancing its complex configuration.
  • CZS-0-1 displayed significant anticoagulant effects similar to heparin by inhibiting specific blood factors (FIIa and FXa), with its activity closely linked to its molecular weight, suggesting potential as a source for bioactive polys

We present the application of -difluoroacetylglucosamine (GlcNDFA) in a chemical evolution strategy to synthesize oligosaccharides. In comparison to conventional -trifluoroacetylglucosamine, GlcNDFA exhibits superior substrate compatibility with glycosyltransferases as well as stability in aqueous environments. Using our 16-step assembly line, GlcNDFA can be used to produce homogeneous dekaparin, a heparin-like medication, with a yield of 62.

P-selectin has been shown to enhance growth and metastasis of mouse tumors by promoting regulatory T cell (Treg) infiltration into the tumors. Theoretically, a P-selectin antagonist could suppress the process. Popylene glycol alginate sodium sulfate (PSS) is a heparin-like marine drug, which was originally approved to treat cardiovascular disease in China.
