The high stereo- and substrate specificities of enzymes have been utilized for micro-determination of amino acids. Here, I review the discovery of l-Phe dehydrogenase and its practical use in the diagnosis of phenylketonuria in more than 5,400,000 neonates over two decades in Japan. Screening and uses of other selective enzymes for micro-determination of amino acids have also been discussed.

An analytical method for the determination of sialic acids in biological samples has been developed and applied to fetal bovine serum (FBS), newborn calf serum and adult bovine serum. The hydrolysis of sera was carried out and the liberated sialic acids were quantified using a rapid and sensitive HPLC. The HPLC includes the separation and detection of N-acetylneuraminic acid (Neu5Ac) and N-glycolylneuraminic acid (Neu5Gc) using hydrophilic interaction liquid chromatography and a fluorometric post-column reaction with 2-cyanoacetamide.

Plackett-Burman and Box-Behnken designs as chemometric tools for micro-determination of l-Ornithine.

Plackett-Burman (PB) and Box-Behnken (BB) screening and response surface factorial designs were used to evaluate spectrophotometric and spectrofluorimetric approaches for the determination of l-Ornithine (ORN) as per se and in dietary supplements. Both approaches were based on the derivatization of the primary amino group of ORN via Hantzsch condensation reaction producing yellow coloured adducts (dihydrolutidine derivative). The reaction product was determined spectrophotometrically (method A) at λ=327nm and spectrofluorimetrically (method B) at 480nm (λ) after excitation at 325nm (λ).

Hydrogen peroxide (HO) is a major antibacterial activity-associated biomarker in honey. Measurement of endogenous HO in honey is of great value in prediction of the HO-depended antibacterial activity and characterization or selection of honey samples for their use as an antibacterial agent or natural food preservative. Considering current methods for HO determination are either time-consuming or complicated with their high-cost, a study was conducted to modify and validate the spectrophotometry-based ferrous oxidation-xylenol orange (FOX-1) method for micro-determination of HO in honey samples.
