measurement and mapping of oxygen levels within the tissues are crucial in understanding the physiopathological processes of numerous diseases, such as cancer, diabetes, or peripheral vascular diseases. Electron paramagnetic resonance (EPR) associated with biocompatible exogenous spin probes, such as Ox071 triarylmethyl (TAM) radical, is becoming the new gold standard for oxygen mapping in preclinical settings. However, these probes do not show tissue selectivity when injected systemically, and they are not cell permeable, reporting oxygen from the extracellular compartment only.

Recent investigations into radiation-induced side effects have focused on understanding the physiopathological consequences of irradiation on late-responding tissues like the spinal cord, which can lead to chronic progressive myelopathy. Proton therapy, an advanced radiation treatment, aims to minimize damage to healthy tissues through precise dose deposition. However, challenges remain, particularly regarding the variation in dose distribution, characterized by maximum deposition at the end of the proton range, known as the distal fall-off of a spread-out Bragg peak.

The neurovascular unit (NVU) is highly responsible for cerebral homeostasis and its dysfunction emerges as a critical contributor to Alzheimer's disease (AD) pathology. Hence, rescuing NVU dysfunction might be a viable approach to AD treatments. Here, we fabricated a self-regulated muti-functional nano-modulator (siR/PIO@RP) that can intelligently navigate to damaged blood-brain barrier and release therapeutical cargoes for synergetic AD therapy.
