Ova were sought in the washings of the excised reproductive tracts of 48 patients after they had ovulated. From 24 patients using no contraceptives, two unfertilized ova were recovered. From 24 patients using intrauterine coils, one unfertilized and one fertilized pronuclear ovum were recovered. The latter ovum was at the earliest stage of development recorded for the human.

Using FRET to Define Cdk1-Dependent Ordering of Events During Exit from Second Meiotic M-Phase in Oocytes.

Methods Mol Biol

November 2024

Christopher Chen Oocyte Biology Research Laboratory, UQ Centre for Clinical Research, Herston, QLD, Australia.

Exit from M-phase requires a precise sequence of molecular events for successful completion, with errors in the process resulting in cell death or aneuploidy, a characteristic feature of cancer and the leading cause of pregnancy failure. Exit from the second meiotic division (MII) in oocytes is a unique event triggered by sperm, involving female anaphase II as well as both male and female pronuclear formation. Very little is known about how these events involving two distinct cell types are coordinated.

Institute of Biotechnology, National Taiwan University, Taipei, Taiwan, Republic of China; Centre for Developmental Biology and Regenerative Medicine, National Taiwan University, Taipei, Taiwan, Republic of China; Agricultural Biotechnology Research Centre, Academia Sinica, Taipei, Taiwan, Republic of China. Electronic address:

Research Question: Can spermatozoa penetrate maturing metaphase I (MI) oocytes, and render subsequent development following conventional IVF in a mouse model?

Design: ICR mice were used in this study. Metaphase II (MII) cumulus-oocyte complexes (COC) harvested 15 h after injection of human chorionic gonadotrophin (HCG) were used for IVF as the control group (Group 1). In the treatment group (Group 2), maturing MI COC harvested 7 h after HCG injection were used for IVF.

In vivo fertilization improved the cryotolerance and developmental ability of vitrified-warmed rat fertilized oocytes.

Sci Rep

October 2024

Division of Reproductive Engineering, Center for Animal Resources and Development, Kumamoto University, 2-2-1 Honjo, Chuo-ku, Kumamoto, 860- 0811, Japan.

The cryopreservation of rat embryos is useful for efficiently archiving rat resources in bioresource repositories. The cryopreserved fertilized oocytes can be quickly reanimated to rats with homozygous mutations using embryo transfer. In addition, cryopreserved rat fertilized oocytes are easier to transport than live animals.

Purpose: We aimed to identify the correlation between morphological pronuclear (PN) status and the genetically determined ploidy configuration in preimplantation embryos.

Methods: A retrospective observational study was conducted on 1982 embryos displaying normal fertilization and 380 embryos showing an atypical PN pattern, tested for aneuploidies and ploidy status via preimplantation genetic testing (PGT) between May 2019 and May 2024. Ploidy prediction was performed using a validated targeted-NGS approach and a proprietary bioinformatic pipeline analyzing SNPs B-allele frequency information.
