Palbociclib is a cyclin-dependent kinase 4/6 inhibitor and a commonly used antitumor drug. Many cancers are susceptible to palbociclib resistance, however, the underlying metabolism mechanism and extent of resistance to palbociclib are unknown. In this study, LC-MS metabolomics was used to investigate the metabolite changes of colorectal cancer SW620 cells that were resistant to palbociclib.

Despite the potentialities of electrochemical sensors, these devices still encounter challenges in devising high-throughput and accurate drug susceptibility testing. The lack of platforms for providing these analyses over the preclinical trials of drug candidates remains a significant barrier to developing medicines. In this way, ultradense electrochemical chips are combined with machine learning (ML) to enable high-throughput, user-friendly, and accurate determination of the viability of 2D tumor cells (breast and colorectal) aiming at drug susceptibility assays.

This Review delves into the mechanisms behind drug resistance in colorectal cancer (CRC), particularly examining the role of nutrient depletion and its contribution to multidrug resistance (MDR). The study highlights metabolic adaptations of cancer cells as well as metabolic adaptations of cancer cells under low nutrient availability, including shifts in glycolysis and lipid metabolism. It emphasizes the significance of MDR1 and its encoded efflux transporter, P-glycoprotein (P-gp/B1), in mediating drug resistance and how pathways such as HIF1α, AKT, and mTOR influence the expression of P-gp/B1 under limited nutrient availability.
